子宫蓄脓对犬卵巢和子宫血液学、组织结构及相关基因的影响

【摘要】:子宫蓄脓是临床上常见的中老年犬的产科疾病,不仅对生殖系统造成损伤,还对病犬其他器官造成危害,严重时可引起病犬死亡。该研究病例来源于2012~2014年在扬州大学动物医院就诊犬,其中闭合型子宫蓄脓15例,开放型子宫蓄脓23例。本文以施行绝育术的18例健康母犬作为对照组,对子宫蓄脓病例的临床资料(发病年龄、品种、初次就诊体温、发病季节、发病距离最后一次发情结束间隔、生育史等)、血液学指标、性激素变化规律、子宫与卵巢的病理组织学变化和子宫与卵巢相关基因和蛋白的表达进行研究。研究的结果如下:子宫蓄脓的平均发病年龄为8.63±2.95岁,7岁以上发病率高达84.21%,其中博美犬和京巴犬为本试验中的高发品种,发病率分别为23.68%和18.42%;闭合型子宫蓄脓和开放型子宫蓄脓初次就诊体温分别为39.07±0.99℃和39.03±0.81℃;大部分病例发病是在发情之后2个月内:有生育史的犬占总发病数的21.11%,明显低于无生育史犬的比例。血液指标和激素检测结果表明:闭合型子宫蓄脓组的血液白细胞、单核细胞、嗜中性粒细胞和淋巴细胞的数量高于对照组,且差异极显著(P0.01)。闭合型和开放型子宫蓄脓组的血液平均红细胞容积低于对照组,且差异显著(P0.05)。闭合型和开放型子宫蓄脓组的血液平均血小板容积低于对照组,且极差异显著(P0.01)。外周血及子宫血血清中,雌激素不论在外周血清还是子宫血清中都表现为闭合组最高、开放组次之,对照组最低,且在子宫血清中,闭合组极显著高于对照组(P0.01)。孕酮在外周血清及子宫血清中的变化相似,闭合组均显著高于对照组(P0.05)。子宫蓄脓犬子宫与卵巢的病理组织学变化:子宫蓄脓多伴随CHE的发生,子宫内膜固有层增厚,子宫腺体增大扩张,当腺体极度扩张时,腺体上皮破坏,腺体内充满白细胞及蛋白。子宫内膜上皮周围聚集大量白细胞,严重者出现上皮坏死、脱落,有时可见子宫肌层血管充血扩张。蛋白检测发现:健康犬卵巢、子宫体肌层ER的表达强度较低:子宫蓄脓犬在子宫体内膜及子宫角腺体基质的表达升高。健康犬子宫角和子宫体肌层及腺体PR表达强度较低,卵巢及子宫体内膜表达较高。TLR2及TLR4在子宫蓄脓犬子宫体、子宫角及卵巢中表达均高于健康犬,但差异不显著。基因表达检测分析表明:与对照组相比,TLR2基因在患开放型子宫蓄脓犬卵巢组织中的表达量显著升高(P0.05),而在闭合型子宫蓄脓犬则极显著升高(P0.01);在子宫角组织中,闭合组极显著高于对照组(P0.01)。TLR4基因在闭合型子宫蓄脓犬的子宫角及卵巢中的表达量均高于对照组,差异极显著(P0.01);在卵巢组织中,开放型子宫蓄脓犬的表达量较对照组显著升高(P0.05)。ER-α基因的表达量,在开放型子宫蓄脓犬的子宫角组织中,显著高于对照组(P0.05),而闭合型子宫蓄脓犬极显著高于对照组(P0.01)。PR基因的表达量,在开放型子宫蓄脓犬的子宫角组织中,显著高于对照组(P0.05),而闭合型子宫蓄脓犬极显著高于对照组(P0.01)。结论:临床上,犬子宫蓄脓可引起病犬血液WBC数量、雌二醇及孕酮水平的升高。子宫内膜的病理变化以CHE为主,卵巢为持久黄体或卵巢囊肿。TLR2、TLR4、ER、PR蛋白阳性表达及基因的相对表达量在两组子宫蓄脓犬生殖器官中均有不同程度的升高。
[Abstract]:Uterine sepsis is a clinically common obstetric disease of the middle-aged and old dog. It not only causes damage to the reproductive system, but also causes harm to other organs of the sick dog. It can cause the death of the sick dog in serious case. The case of this study was found in 2012~2014 years in the animal hospital of Yangzhou University, including 15 cases of closed uterine purulent and 23 cases of open type of uterus. The clinical data (age, variety, body temperature, season, distance of the last estrus, birth history, etc.), Hematology Index, sex hormone change law, histopathological changes of uterus and ovary, and uterus and egg of uterus and ovum were used in 18 healthy female dogs as control group. The results of the expression of nests related genes and proteins are studied. The results of the study are as follows: the average age of the onset of uterine sepsis is 8.63 + 2.95 years, and the incidence of the disease is up to 84.21% above the age of 7. The BBO and Beijing Ba dogs are high incidence in this test, with the incidence of 23.68% and 18.42%, respectively. The body temperature was 39.07 + 0.99 C and 39.03 + 0.81 C respectively. Most cases occurred within 2 months after the estrus: the dogs with a history of birth accounted for 21.11% of the total incidence, obviously lower than the proportion of the non bearing history dogs. The blood indexes and hormone test results showed that the blood leukocytes, monocytes and neutrophils in the closed uterine purulent group The number of cells and lymphocytes was significantly higher than that in the control group (P0.01). The blood average blood volume of the closed and open uterine purulent groups was lower than that of the control group, and the difference was significant (P0.05). The blood average platelet volume in closed and open uterine purulent groups was significantly lower than that in the control group (P0.01). In the blood serum of the uterus, estrogen was the highest in the peripheral blood serum and in the uterus serum, the open group and the control group were the lowest, and in the uterine sera, the closed group was significantly higher than the control group (P0.01). The changes of progesterone in the peripheral serum and the uterus serum were similar, the closed group were significantly higher than the control group (P0.05). The uterus was significantly higher than the control group. The histopathological changes in the uterus and ovary of the purulent dog: the uterine sepsis is associated with the occurrence of CHE, the thickening of the inner lamina of the endometrium, the enlargement of the uterus gland, the destruction of the gland epithelium and the white cells and protein in the gland when the gland is extremely dilated. The expression of ER in the healthy canine ovary and the myometrium of the uterus was lower, and the expression of the endometrium and the uterine horn gland matrix in the uterus increased. The expression of the uterus angle and the myometrium and the gland PR in the healthy dog was lower, the expression of the ovary and the endometrium was higher.TLR2. The expression of TLR4 in the uterine body, uterus angle and ovary of the uterus was higher than that of the healthy dog, but the difference was not significant. The analysis of gene expression analysis showed that compared with the control group, the expression of TLR2 gene in the ovary tissue of the open type uterine purulent dog was significantly higher (P0.05), while in the closed uterine purulent dog (P0.01); In the uterine horn tissue, the closed group was significantly higher than the control group (P0.01).TLR4 gene expression in the uterus angle and ovary of the closed uterine purulent dog was higher than the control group, the difference was very significant (P0.01); in the ovarian tissue, the expression of the open type uterus purulent dog increased significantly (P0.05) the expression of.ER- alpha gene, in the opening of the ovary. The uterus angle tissue of the canine type uterine septic was significantly higher than that of the control group (P0.05), while the closed uterine purulent dog was significantly higher than the control group (P0.01).PR gene expression, significantly higher than the control group (P0.05) in the uterus angle tissue of the open type uterus purulent dog (P0.05), and the closed uterine purulent dog was significantly higher than that of the control group (P0.01). In clinical, the Canine Uterus purulent can cause the number of WBC, the level of estradiol and progesterone. The pathological changes of the endometrium are mainly CHE, the ovary is the persistent corpus luteum or ovarian cyst.TLR2, the TLR4, ER, PR protein positive expression and the relative expression of the gene in the two groups of uterus purulent canine reproductive organs have different degrees of increase.
