DON暴露对雏鸡神经递质及钙调蛋白含量的影响

【摘要】:【目的】通过DON暴露体内试验,根据雏鸡脑组织中神经递质含量及钙调蛋白(CAM)含量的变化,研究DON对雏鸡的神经毒性作用。【方法】选取120只健康1日龄海兰蛋鸡(公)预饲1周后,按单因素试验设计随机均分为DON低、中、高剂量组和对照组,对照组和试验组饲喂相同的全价日粮。DON低剂量组、中剂量组和高剂量组分别按采食量中含DON 0.27,1.68和12.21mg/kg的剂量灌胃,对照组灌服等量的生理盐水。试验从第8天开始,每隔7d染毒1次,共染毒5次。试验期为36d。试验结束时,每组随机屠宰20只鸡,迅速取出脑组织,检测神经递质含量和CAM含量的变化。【结果】经高效液相色谱-荧光检测法(HPLC-FD)检测,雏鸡脑组织中未发现5-羟色胺吲哚乙酸(5-HIAA);与对照组相比,DON高剂量组雏鸡脑组织中去甲肾上腺素(NE)和5-羟色胺(5-HT)含量显著升高(P0.05),而多巴胺(DA)含量显著降低(P0.05)。与对照组相比,DON中、高剂量组雏鸡神经细胞内游离钙([Ca2+]i)浓度和脑组织中CAM含量显著降低(P0.05),不同DON剂量组CAM mRNA基因相对表达量均显著降低(P0.05)。【结论】DON暴露可引起雏鸡神经钙稳态变化,并影响部分神经递质的分泌,对雏鸡具有一定的神经毒性作用。
[Abstract]:[objective] to study the changes of neurotransmitters and calmodulin (calmodulin) contents in brain tissue of chicks by DON exposure in vivo. The neurotoxic effects of DON on chicks were studied. [methods] 120 healthy 1-day-old Hailan laying hens (males) were prefed for 1 week, and were randomly divided into low, middle, high dose DON groups and control groups according to the single factor design. The control group and the experimental group were fed with the same low-dose diet. The middle dose group and the high dose group were fed with the same dose of DON 0.27N 1.68 and 12.21mg/kg, respectively. The control group was given the same amount of normal saline. The experiment began on the 8th day and was administered once every 7 days for a total of 5 times. The experimental period is 36 days. At the end of the experiment, 20 chickens were slaughtered at random in each group, the brain tissue was removed quickly, and the changes of neurotransmitter and CAM content were detected. [results] HPLC-FDD was used to detect the content of neurotransmitter and CAM. Compared with the control group, the contents of NE) and 5-HT) in brain tissues of chicks with high dose of don were significantly higher than those of the control group, while the contents of dopamine (DA) decreased significantly in the brain tissues of chicks. In don compared with the control group, The concentration of free calcium ([Ca2] I) in nerve cells and the content of CAM in brain tissue decreased significantly in high dose group, and the relative expression of CAM mRNA gene decreased significantly in different DON dosage groups. [conclusion] DON exposure can induce steady state change of nerve calcium in chicks. It also affected the secretion of some neurotransmitters and had a certain neurotoxic effect on chicks.
