一株凝结芽孢杆菌的分离鉴定及益生性研究

【摘要】:试验旨在分离鉴定一株来源于池塘底泥的具有益生功能的凝结芽孢杆菌,并研究其益生性能。从自然池塘底泥分离得到一株产酸能力强的杆状细菌,通过生理生化特征分析和16SrDNA序列比对,鉴定分离菌为凝结芽孢杆菌(Bacillus coagulans)。该分离菌株对大肠杆菌K88、大肠杆菌K99、无乳链球菌和嗜水气单胞菌有抑制作用;在pH 2.5的模拟胃液中处理12h存活率为71.9%;在胆盐浓度为0.3%的模拟肠液中处理12h,存活率为84.4%。该分离菌株可有效降解亚硝酸盐,48h降解率达到96.80%。将270只1日龄AA肉鸡分成3个处理组,日粮分别添加0(Ⅰ组)、1.0×10~6(Ⅱ组)和1.0×10~7 CFU/g(Ⅲ组)剂量的凝结芽孢杆菌NJ102,试验期42d。结果显示,Ⅱ、Ⅲ组肉鸡的平均日增重和终末体重显著高于Ⅰ组(P0.05),料重比显著低于Ⅰ组(P0.05),各组间平均日采食量差异均不显著(P0.05)。结果表明,来源于池塘底泥的凝结芽孢杆菌NJ102有一定的抗逆性和益生能力,能改善肉鸡的生长性能,具备作为微生物饲料添加剂开发的潜力。
[Abstract]:The purpose of this study was to isolate and identify a probiotic Bacillus coagulans from pond sediment and to study its probiotic properties. A rod-like bacterium with strong acid-producing ability was isolated from the sediment of natural pond. The bacteria was identified as Bacillus coagulansa by physiological and biochemical analysis and 16SrDNA sequence alignment. The strain had inhibitory effect on Escherichia coli K88, Escherichia coli K99, Streptococcus hydrophila and Aeromonas hydrophila, and the survival rate was 71.9 in simulated gastric juice at pH 2.5 and 84.4in simulated intestinal fluid containing 0.3% bile salt for 12 hours.
