猪不同泌乳期母乳外泌体中let-7a和let-7f的差异表达研究

【摘要】:【目的】为研究miRNAs在仔猪免疫系统发育过程中的调控作用。【方法】本研究采用荧光定量PCR技术(Q-PCR)研究了不同泌乳期(0、3、7、14、21和28 d)的梅山猪和大约克夏猪母乳外泌体中let-7a和let-7f的差异表达。【结果】let-7a和let-7f在泌乳早期(0~3 d)的表达丰度明显高于泌乳后期(7~28 d);品种间分析显示梅山猪母乳外泌体中let-7a和let-7f在整个泌乳期(0~28d)的表达水平均明显高于大约克夏猪。【结论】研究表明,let-7a和let-7f能够正向调控仔猪免疫系统发育。本研究可为阐明母乳外泌体包被的miRNA在幼畜免疫系统发育过程中的具体调控机制提供基础数据。
[Abstract]:[objective] to study the regulatory role of miRNAs in the development of immune system in piglets. [methods] in this study, the milk exocrine of Meishan pig and large Yorkshire pig at different lactation stages were studied by fluorescence quantitative PCR technique and Q-PCR. [results] the expression abundance of let-7a and let-7f in the early lactation stage was significantly higher than that in the late lactation stage (728 days), and the expression level of let-7a and let-7f in the breast milk exocrine of Meishan pig during the whole lactation period was significantly higher than that in the late lactation stage at 728 d). [results] the expression level of let-7a and let-7f in the breast milk exocrine of Meishan pig was significantly higher than that in the late lactation stage. [conclusion] the results showed that let-7a and let-7f could positively regulate the development of immune system in piglets. This study can provide basic data for elucidating the regulation mechanism of miRNA in the development of immune system in young animals.
