妊娠后期宫内生长限制对蒙古绵羊胎儿胸腺微环境的影响

【摘要】:本实验旨在研究妊娠后期宫内生长限制对蒙古绵羊胎儿胸腺微环境的影响。选取同期发情受孕(B超确定怀单羔)的健康蒙古绵羊24只,当蒙古绵羊妊娠期达到90d时,挑选出与总体体重平均值相近的6只怀孕母羊解剖取胎儿及其胸腺,设为90d组,剩下的18只按体重随机分成不同营养水平组,共三组:0.18 MJME·BW-0 75·d-1(RG1:n=6)、0.33 MJME·BW-0.75·d-1(RG2:n=6)和 0.67 MJME·BW-0.75·d-1(CG:n=6),饲养到妊娠 140d时每组全部屠宰。试验结果表明:(1)妊娠后期母体受营养限制,导致RG1组胎儿重量、胸腺重量相较于CG组均极显著降低(p0.01);同时RG2组胎儿重量比CG组极显著降低(p0.01),而胸腺重量则显著低于CG组(P0.05)。随着限制程度的加深,胸腺中的胶原纤维和网状纤维逐渐增多。(2)妊娠后期RG1组胎儿胸腺中Ⅰ型胶原、纤连蛋白含量均极显著低于CG组(P0.01),层粘连蛋白、透明质酸含量则都显著高于CG组(P0.05);RG2组胎儿胸腺中仅有纤连蛋白含量极显著低于CG组(P0.01)。RG1组胎儿胸腺激素中胸腺生成素、胸腺肽β4均显著高于CG组(P0.05);炎性因子中TNF-a(P0.01)极显著低于CG组,IL1β、IL4均显著低于CG组(P0.05);趋化因子中CCL25(P0.01)极显著低于CG组,CXCL10、CXCL12、β 2微球蛋白均显著低于CG组(P0.05);生长因子中EGF、EGFR、KGF、NGF、NGFR、FGFR3 均显著低于 CG 组(P0.05),FGFR2(P0.05)显著高于CG组。RG2组胎儿胸腺炎性因子中TNF-a(P0.01)极显著低于CG组,IL1 β、IL4显著低于CG组(P0.05);趋化因子中CXCL10、CXCL12显著低于CG组(P0.05);生长因子中EGF、EGFR均显著低于CG组(P0.05),FGFR2(P0.05)显著高于CG组。(3)妊娠后期RG1、RG2组胎儿胸腺EMT转化指标中E-cad、PCDH8均显著低于CG组(P0.05),RG1组中OB-cadherin(P0.05)显著高于CG组。
[Abstract]:The purpose of this study was to study the effect of intrauterine growth restriction on thymus microenvironment in Mongolian sheep. Twenty-four healthy Mongolian sheep were selected for simultaneous estrus conception (B ultrasound to determine single lamb). When the gestation period of Mongolian sheep reached 90 days, 6 pregnant ewe with similar average body weight were selected to dissect the fetus and its thymus, which were divided into 90 days group. The remaining 18 rats were randomly divided into three groups according to their body weight. Three groups: 0.18 MJME BW-0 75 d ~ (-1) RG1: n ~ (1) (0.33 MJME BW-0.75 d ~ (-1) RG2: n ~ (6) and 0.67 MJME BW-0.75 d ~ (-1) CG1: n ~ (6) were raised and slaughtered in each group until day 140 gestation. The results showed that the fetal weight and thymus weight of RG1 group were significantly lower than those of CG group, and the fetal weight of RG2 group was significantly lower than that of CG group, while the weight of thymus was significantly lower than that of CG group. With the deepening of restriction, collagen fibers and reticular fibers in thymus increased gradually. (2) Type I collagen and fibronectin content in fetal thymus in RG1 group were significantly lower than those in CG group (P 0.01 and laminin). The content of hyaluronic acid in fetal thymus of CG group was significantly higher than that of CG group. Only fibronectin content in fetal thymus of CG group was significantly lower than that of CG group (P 0.01). RG1 group was significantly lower than that of CG group. Thymosin 尾 4 was significantly higher than CG group (P 0.05), inflammatory factor TNF-a (P 0.01) was significantly lower than CG group (P 0.05), chemotactic factor CCL25 (P 0.01) was significantly lower than CG group (CXCL10CXCL12), 尾 2 microglobulin was significantly lower than CG group (P 0.05), and EGFEGFRNGFNGFNGFNGFR3 in CG group was significantly lower than that in CG group (P < 0.05). FGFR2P0.05) was significantly higher than that of CG group. RG2 group (TNF-aFGFR2P0.01) was significantly lower than CG group (P 0.05); CXCL10CXCL12 was significantly lower than CG group (P0.05); and growth factor (EGFR) was significantly lower than CG group (P0.05FGFR2FGFR2 P0.05). The level of EGFR in CG group was significantly lower than that in CG group (P < 0.05); the level of CXCL10CXCL12 in CG group was significantly lower than that in CG group (P < 0.05); the level of EGFR in growth factor was significantly lower than that in CG group (P < 0.05); and that in CG group was significantly lower than that in CG group. The levels of E-cad-PCDH8 in fetal thymus of RG1 / RG2 group were significantly lower than those of CG group (P0.05 / RG1 group) and significantly higher than that of CG group (P < 0.05).
