12个lncRNA在发生炎症反应仔猪骨骼肌和肝脏组织中的表达分析

【摘要】:为探讨12个长链非编码RNA(lnc RNA)在发生炎症反应的仔猪骨骼肌和肝脏组织炎症反应中的差异表达,本研究选取12头(28±3)日龄、体重(9.0±0.7)kg的杜×长×大断奶仔猪,分成2个处理,每个处理6个重复;2个处理组基础饲粮相同,试验第21天,LPS组腹膜注射100μg/kg体重(BW)的LPS,对照组注射等量的生理盐水。于注射LPS或生理盐水4 h后屠宰,取骨骼肌和肝脏样品待测。结果表明:LPS刺激导致骨骼肌和肝脏组织中肿瘤坏死因子-α(TNF-α)、环氧合酶2(COX2)和核苷酸结合寡聚域受体2(NOD2)的m RNA表达量显著提高(P0.05);LPS刺激极显著降低骨骼肌中lnc RNA-36199.1的表达量(P0.01);LPS刺激显著提高肝脏中lnc RNA-MEG3、lnc RNA-RP11-451G4.2、lnc RNA-27609.1、lnc RNA-32021.1、lnc RNA-46606.2和lnc RNA-36199.1的表达量(P0.05),但显著降低lnc RNA-30795.1和lnc RNA-26244.1的表达量(P0.05)。本试验研究表明,8种lnc RNA在LPS诱导的断奶仔猪骨骼肌或肝脏的炎症反应中存在差异表达,说明其可能参与不同组织的炎症反应。
[Abstract]:In order to investigate the differential expression of 12 long chain noncoding RNA(lnc RNAs in skeletal muscle and liver tissues of piglets with inflammatory response, 12 piglets with 9. 0 卤0.7)kg body weight and 28 卤3 days of age were selected and divided into 2 treatments. Two treatment groups received the same basal diet. On the 21st day of the experiment, LPSs of 100 渭 g/kg BW were injected into the peritoneum of LPS-treated group and the control group was injected with the same amount of normal saline. Four hours after injection of LPS or normal saline, skeletal muscle and liver samples were slaughtered. The results showed that the expression of m RNA of TNF- 伪 TNF- 伪, cyclooxygenase-2 (COX2) and nucleotide-binding oligodeoxyribonucleotide receptor (2nd2) in skeletal muscle and liver was significantly increased by the stimulation of TNF- 伪 TNF- 伪, COX-2) and the expression of lnc RNA-36199.1 in skeletal muscle by the stimulation of lipopolysaccharide (LPS). The expression of lnc RNA-MEG3LNC RNA-RP11-451G4.2LNC RNA-27609.1lncRNA-32021.1lnc RNA-46606.2 and lnc RNA-36199.1 in liver was significantly increased by LPS stimulation, but the expression of lnc RNA-30795.1 and lnc RNA-26244.1 was significantly decreased (P0.05). This study showed that eight lnc RNA were differentially expressed in skeletal muscle or liver of weaned piglets induced by LPS, suggesting that they may participate in the inflammatory response of different tissues.
