羊口疮病毒感染后皮肤成纤维细胞及病毒转录组学分析

【摘要】:羊口疮病毒(orfv virus;ORFV)可引起山羊和绵羊的一种急性接触性传染病,也可感染人、牛和其它脊椎动物,是一种自限性疾病。由于其安全性高、短期内可反复感染并且具有强大的免疫调节活性,因此可作为一种安全有效的病毒载体用于多种感染性疾病以及肿瘤治疗研究,目前已在人类肿瘤治疗模型中取得初步的治疗效果。然而ORFV感染人体细胞后引起宿主细胞基因的转录调控以及病毒与细胞之间的相互作用却研究得较少。我们发现ORFV可在皮肤成纤维细胞中复制增殖并正常表达病毒蛋白,因此可通过mRNA测序技术来研究人皮肤成纤维细胞在ORFV感染后细胞及病毒基因转录表达水平的特异性变化,进而深入了解ORFV对人细胞的转录调控以及二者之间的相互作用。收集ORFV(MOI=5)感染后0、3和8小时的人皮肤成纤维细胞,抽提RNA并纯化,质检合格(RIN9.5,28S/18S=1.6)后再进行测序文库构建、簇生成和上机测序。原始测序结果经接头、测序引物、核糖体RNA和低质量序列的过滤后,与人基因组进行比对。我们将q值(校正后的p值)
[Abstract]:Sheep mouth sore virus orfv virus (ORFV) can cause an acute contact infectious disease in goats and sheep, but also infect humans, cattle and other vertebrates. It is a self-limited disease. Because of its high safety, reproducible infection in the short term and strong immunomodulatory activity, it can be used as a safe and effective viral vector for the study of various infectious diseases and tumor therapy. At present, the primary therapeutic effect has been obtained in human tumor treatment model. However, little research has been done on the transcriptional regulation of host cell genes and the interaction between viruses and cells caused by ORFV infection. We found that ORFV can replicate and proliferate in skin fibroblasts and normally express viral proteins, so we can study the specific changes of transcription expression level of human skin fibroblasts and viral genes after ORFV infection by mRNA sequencing. Furthermore, the transcriptional regulation of ORFV on human cells and the interaction between them are further understood. The human skin fibroblasts were collected for 0 and 8 hours after infection. RNA was extracted and purified. The sequence library was constructed and sequenced. The primordial sequencing results were compared with the human genome by the filter of the connectors, sequencing primers, ribosomal RNA and low quality sequences.
