水貂阿留申病分子检测及其对产仔性能的影响

【摘要】:水貂(Mink)是哺乳纲鼬科鼬属的肉食性动物,主要用以皮毛生产,是世界三大毛皮商品之一,被誉为“裘皮之王”。目前市场上除了黑褐色之外,还出现了各种色泽艳丽的彩色水貂。而水貂除毛皮之外,还可制作许多副产品。可谓是由内而外都具有极高的经济价值。水貂阿留申病是养貂产业公认的三大疫病之一。主要侵害水貂的免疫系统,引发动脉血管炎、肾小球肾炎等疾病。患病水貂消瘦、贫血、衰弱,皮毛质量差。一般对患病母貂造成空怀、流产、弱胎,而公貂精子活力和配种能力降低,造成严重的经济损失。研究表明,阿留申病因其特殊的发病机理,目前还没有安全有效的治疗方法。所以要想做好水貂阿留申病的控制工作,最大程度的降低该病对水貂产业造成的损失,最有效的手段就是在养殖过程中制定严格的疾病检测制度,及时发现并淘汰病貂。因此,国内外学者探究出多种水貂阿留申病的检测手段,在水貂的生产养殖中得以广泛应用。粪便DNA提取方法是非损伤性取样中的一种,该方法在取样过程中避免了对动物个体的干扰和损伤,且检测样本量大、成本低廉,在动物分子生物学研究中具有很高的实际意义和应用价值。水貂作为一种名贵的毛皮经济动物,在对其进行疾病检测过程中,应注意保护它的经济价值,尽量降低对其造成影响。在水貂阿留申病的检测过程中,利用粪便DNA提取的检测方法,既可以避免对水貂个体造成影响和损伤,又能够准确的测定其阿留申病的感染情况,具有很高的使用操作价值。因此,本研究利用水貂粪便DNA提取技术,来检测水貂体内阿留申病毒,并以此为基础,探讨阿留申病对水貂产仔数的影响。主要试验内容结果如下:(1)利用Ezup柱式病毒DNA抽提试剂盒,成功的从患病水貂粪便中,抽提出阿留申病毒DNA。说明该种方法可以用于水貂阿留申病的检测。(2)从产仔数高、低两组粪样中抽提的阿留申病毒DNA经PCR扩增后,电泳结果显示,低产仔数组带毒率高于高产仔数组。说明阿留申病对水貂产仔数有影响,引起水貂产仔数降低。综上所述,本研究证实,病毒DNA抽提试剂盒可以提取水貂粪便中阿留申病毒DNA,该方法可以用以水貂阿留申病的检测;水貂阿留申病影响水貂产仔个数,造成患病水貂产仔数下降。
[Abstract]:Mink (Mink) is a carnivorous animal of the weasel genus of the weasel family. It is mainly used for fur production. It is one of the world's three largest fur goods. It is known as the "king of fur". In addition to the black brown market, there are many colorful colored mink in the market. Sable Aleutian disease is one of the three major diseases recognized by mink industry. It mainly infringes the immune system of mink, causes arterial vasculitis, glomerulonephritis and other diseases. The sable is emaciated, anemic, weak, and poor in fur quality. The study shows that there is no safe and effective treatment for Aleutian disease, so it is necessary to make the control of Aleutian disease and reduce the loss of the mink industry to the greatest extent. The most effective means is to make it in the breeding process. Therefore, domestic and foreign scholars have explored a variety of mink Aleutian disease detection methods, and have been widely used in the production of mink. The method of DNA extraction is one of non damage sampling. This method avoids the interference and damage to animal individuals during the sampling process. It has high practical significance and application value in the study of animal molecular biology. As a valuable animal, mink should protect its economic value and reduce its impact on it in the process of disease detection. In the detection process of Aleutian disease of mink, mink is a kind of expensive fur economic animal. Using the detection method of fecal DNA extraction, it can not only avoid the influence and damage to mink, but also can accurately determine the infection of Aleutian disease. Therefore, this study uses the DNA extraction technique of mink feces to detect Aleutian virus in mink, and based on this, it discusses Aleutian. The main results were as follows: (1) using the Ezup column virus DNA extraction kit, the Aleutian virus DNA. was successfully extracted from the dung of the sable mink. The method could be used to detect Aleutian disease in mink. (2) the Aleutian virus (DNA) from the number of litter and the two groups of feces in the low two groups was expanded by PCR. After the increase, the electrophoresis results showed that the toxicity rate of the low yield array was higher than the high yield array. It showed that Aleutian disease had an impact on the number of mink litter, and the number of mink litter decreased. In summary, this study confirmed that the virus DNA extraction kit could extract Aleutian virus DNA in mink feces; this method can be used for the detection of Aleutian disease in Mink; Mink Aleutian disease affects mink litter size, resulting in a decrease in litter size in the affected mink.
