中药提取物复方制剂抗新城疫病毒的药效及质量控制

【摘要】:鸽新城疫(newcastle disease,ND)又称鸽瘟或鸽Ⅰ型副粘病毒(pigeon paramyxovirus type 1,PPMV-1),是由新城疫病毒(newcastle disease virus,NDV)引起的一种急性、热性、败血性和高度接触性传染病。为预防和治疗该疾病的发生,大量使用抗生素和化学类药物,导致耐药性和药物残留问题的出现。而且随着生态养殖和绿色食品等概念的提出,我国规定在饲料和食品中严禁使用兽医临床中抗生素类药物和化学类药物。中药具有抗病毒作用效果好、毒副作用小和无耐药性等优点,不但能提高机体免疫力,还能促进生长,提高生产性能等。本研究在中兽医药理论指导下,以金银花、黄芩、连翘、贯叶连翘等10余味中药提取物进行中药提取物复方制剂,研究中药提取物复方制剂的抗病毒能力、免疫增强能力和质量控制标准。结果如下:1.中药提取物复方制剂高剂量(含生药2 g/mL)预防、治疗组和中剂量(含生药1 g/mL)预防、治疗组的血凝效价显著降低(P0.01),其中中药提取物复方制剂中剂量组(含生药1 g/m L)的效果显著。2.中药提取物复方制剂能够提高NDV疫苗抗体效价,中药提取物复方制剂中剂量组(含生药1 g/mL)的抗体效价差异显著(P0.05);中药提取物复方制剂中剂量组法氏囊器官指数差异显著(P0.05);免疫器官内淋巴细胞排列紧密,且淋巴细胞数量高于阳性对照组。3.以黄芩苷为检测成分,利用TLC和HPLC检测方法对中药提取物复方制剂进行质量控制研究。TLC结果显示,黄芩苷供试品和标准品在相应的位置上出现了颜色相同的斑点,阴性对照品中无干扰现象;HPLC结果显示,黄芩苷在浓度为10~200μg/mL之间时线性关系良好,黄芩苷的含量为3727.41 mg/g,在供试品中的黄芩苷峰保留时间与标准品中的峰是一致的,且阴性对照品无干扰现象。中药提取物复方制剂对新城疫病毒具有较好的抑制作用,能够显著提高NDV疫苗抗体效价和法氏囊器官指数,免疫器官内淋巴细胞排列紧密且数量增多。所建立的黄芩苷TLC和HPLC检测方法,专属性强,可快速准确地对中药提取物复方制剂中黄芩苷进行检测。
[Abstract]:Pigeon Newcastle disease (NDV), also known as pigeon plague or pigeon paramyxovirus type 1 (PPMV-1), is an acute, feverish, septic and highly contagious disease caused by Newcastle disease virus. In order to prevent and treat the disease, antibiotics and chemical drugs are used extensively, which leads to the emergence of drug resistance and drug residues. With the concept of ecological breeding and green food being put forward, the use of antibiotics and chemical drugs in veterinary clinic is strictly prohibited in feed and food in China. Traditional Chinese medicine has the advantages of good antiviral effect, little side effect and no drug resistance. It can not only improve the body immunity, but also promote growth, improve the production performance and so on. In this study, under the guidance of Chinese veterinary medicine theory, 10 aftertaste extracts such as Flos Lonicerae, Radix Scutellariae, Forsythia Forsythia and Hypericum perforatum were used to prepare Chinese medicine extract compound preparation, and the antiviral ability of compound preparation of traditional Chinese medicine extract compound preparation was studied. Immune enhancement and quality control standards. The result is as follows: 1. High dose (including crude drug 2 g / mL) prevention, treatment group and middle dose (containing crude drug 1 g / mL) prevention, The hemagglutination titer of the treatment group was significantly lower than that of the control group, and the effect of the middle dose group (containing 1 g / m L crude drug) was significant. The compound preparation of Chinese traditional medicine extract can improve the antibody titer of NDV vaccine. The antibody titers of the middle dose group (containing 1 g / mL crude drug) were significantly different from those of the middle dose group (P 0.05), the difference of the bursa organ index was significant (P 0.05), and the lymphocytes in the immune organs were closely arranged. The number of lymphocytes was higher than that of positive control group. Using baicalin as the detection component, the quality control of compound preparation of traditional Chinese medicine extract was studied by TLC and HPLC. The results showed that the same color spots appeared in the samples and standard samples of baicalin. The results of HPLC showed that baicalin had a good linear relationship when the concentration of baicalin was 10 渭 g / mL, and the content of baicalin was 3727.41 mg / g. The retention time of baicalin peak in the sample was the same as that in the standard sample. And negative reference substance has no interference phenomenon. The compound preparation of Chinese traditional medicine extract has a good inhibitory effect on Newcastle disease virus, can significantly improve the antibody titer of NDV vaccine and the bursal organ index of bursa, and the lymphocyte arrangement in immune organs is close and the number of lymphocytes increases. The established TLC and HPLC methods for the detection of baicalin can be used for rapid and accurate determination of baicalin in compound preparations of traditional Chinese medicine.
