2014年中国11省市猪流行性腹泻病毒ORF3基因分子流行病学调查

【摘要】:猪流行性腹泻(porcineepidemicdiarrhea,PED)是由猪流行性腹泻病毒(porcineepidemicdiarrheavirus,PEDV)引起的猪的一种急性、高度传染性的肠道疾病。该病毒主要破坏猪的消化道器官,其主要临床症状为急性肠炎、呕吐、水样腹泻和脱水。各年龄段以及不同品系的猪均对PEDV敏感,该病毒对哺育仔猪危害性非常大,其发病率为100%,死亡率为80~100%,给养猪业造成了严重的经济损失。 2010年末PED在我国华南地区发生,随后在国内广泛流行,发病率和死亡率居高不下。为了研究猪流行性腹泻病毒在中国的流行情况,本课题组于2014年1月至12月对来自9个省份和2个直辖市(黑、吉、辽、内蒙古、冀、鲁、晋、苏、湘、京、津)42个发生腹泻猪场的165份腹泻样品,通过胶体金和RT-PCR等一系列实验方法对其进行了检测,结果显示,有25个猪场仅检测出了PEDV,占总数的59.52%;有11个猪场还检出了传染性胃肠炎病毒(TGEV)和猪轮状病毒(PoRV),占总数的26.19%。165份被检测的病料中,病料总数44.24%的病料单独感染PEDV,数量为73份;有55份PEDV、TGEV、PoRV混感的病料,占总数的33.33%;其余样品的病原为其他肠道病病原。在42个发病猪场中,感染PEDV的猪场有36个,占猪场总数的85.71%;165份病料样品中,77.6%的病料样品呈PEDV阳性,数量为128份。本实验结果表明:PEDV在我国的猪腹泻病病原中占主导地位。 根据获得的21株ORF3基因检测结果表明,均由675个核苷酸组成,与GenBank上已登录的12株国内外参考毒株进行序列比对,同源性为90.6%~100%。与国内外毒株同源性比较结果显示,,中国目前流行毒株与欧洲毒株和中国较早流行毒株同源性较低,而与韩国毒株同源性较高。33个PEDV毒株的ORF3基因系统发育树显示,中国PEDV现地野毒株和PEDV参考毒株被分为2个群。
[Abstract]:Porcineepidemicdiarrhea (PED) is an acute, highly contagious intestinal disease caused by porcineepidemicdiarrheavirus (PEDV). The virus mainly destroys the digestive tract organs of the pig. Its main clinical symptoms are acute enteritis, vomiting, water like diarrhea and dehydration. The pigs of different strains were sensitive to PEDV, and the virus was very harmful to the piglets, the incidence was 100% and the mortality rate was 80~100%, which caused serious economic loss to the pig industry.
At the end of 2010, PED took place in the Southern China area of China, followed by widespread prevalence in China, and the incidence and mortality rate remained high. In order to study the prevalence of porcine epidemic diarrhea virus in China, the subject group took 42 abdomen from January 2014 to December to 9 provinces and 2 municipalities (black, Ji, Liao, Inner Mongolia, Hebei, Lu, Jin, Su, Xiang, Jing, Tianjin). 165 samples of diarrhoea in diarrhea piggery were tested by a series of experimental methods such as colloid gold and RT-PCR. The results showed that only 25 pig farms were detected only PEDV, accounting for 59.52% of the total, and 11 piggery also detected infectious gastroenteritis virus (TGEV) and porcine rotavirus (PoRV), which accounted for the total number of 26.19%.165 samples. A total of 44.24% of the total diseases were infected with PEDV, with a number of 73 copies, 55 PEDV, TGEV, and PoRV mixed diseases, accounting for 33.33% of the total; the other samples were pathogenic to other intestinal diseases. In 42 swine farms, 36 pigs infected with PEDV, 85.71% of the total pig farm, and 77.6% of the 165 samples of the disease samples were PEDV positive. Sex and quantity were 128 copies. The results showed that PEDV played a leading role in the pathogenesis of swine diarrhea in China.
According to the results of the detection of 21 ORF3 genes, 675 nucleotides were made up and compared with the 12 domestic and domestic reference strains on GenBank. Homology from 90.6% to 100%. and homology of domestic and foreign strains showed that the homology of the current epidemic strains in China and the early epidemic strains of Europe and China were low. The ORF3 gene phylogenetic tree of.33 PEDV strains with high homology with the Korean strain showed that the Chinese PEDV wild strain and PEDV reference strain were divided into 2 groups.
