猪胸膜肺炎放线杆菌的分离鉴定及其生物学特性的研究

【摘要】:为了对疑似猪接触传染性胸膜肺炎的病死猪肺脏进行胸膜肺炎放线杆菌(APP)的分离鉴定,并研究其生物学特性,通过形态学及培养特性观察、生化试验、PCR方法对分离菌株进行了鉴定,并对APP分离株进行了NAD依赖试验生物学分型、PCR方法血清学分型、PFGE基因分型以及K-B纸片法药敏试验。结果显示,共分离到16株APP,源自两省三地的APP分离株具有一致的形态学特征和相似的生化特性。9株福建分离菌均为生物Ⅱ型,其PFGE基因型均为Pa4;7株安徽分离菌均为生物I型,其中6株为血清型12型,菌株FD1402的PFGE基因型为Pa2,其余5株APP菌株的PFGE基因型为Pa1,剩余1株为血清型3型,其PFGE基因型为Pa3。药敏试验结果显示,对18种常用抗菌药中的环丙沙星、诺氟沙星、氟苯尼考,安徽株均100%敏感,福建株依次为88.9%、77.8%和66.7%敏感;所有分离株对其他15种药物均表现出不同程度的耐药。上述结果表明,不同来源APP菌株的生物学特性较为一致。血清型12型是安徽省APP的主要流行血清型之一。APP分离株的PFGE基因型与血清型、生物型之间无明显的相关性。
[Abstract]:In order to isolate and identify Actinobacillus pleuropneumoniae (app) from the lungs of pigs suspected to be exposed to infectious pleural pneumonia, and to study its biological characteristics, morphological and cultural characteristics were observed. Biochemistry test and PCR method were used to identify the isolated strains, and NAD dependent biological typing was carried out for app isolates. The serological typing was performed by PCR method, and the PFGE genotyping and K-B disk method drug sensitivity test were also carried out. The results showed that 16 APP strains were isolated from the two provinces and three regions. The 9 strains isolated from Fujian were biotype 鈪,
