比色法测定比格犬尿液中柠檬酸的浓度

【摘要】:建立了一种测定比格(Beagle)犬尿液中柠檬酸浓度的方法,利用柠檬酸与溴化钾、高锰酸钾在酸性溶液中生成五溴丙酮,五溴丙酮经石油醚(90℃~120℃)萃取后,与硫脲反应生成水溶性黄色复合物,其用紫外分光光度计在435nm波长处测定。结果显示,柠檬酸浓度在0.005mg/mL~0.25mg/mL范围内,水溶性黄色复合物的吸光度与柠檬酸的浓度呈良好的线性关系,OD435=1.8335C+0.0038,r=0.9996,平均回收率为99.954%(n=5),变异系数为1.64%(n=20)。该方法操作简单,快速准确,适用于非临床研究中Beagle犬尿液中柠檬酸浓度的测定。
