紫外线b促进绵羊黑色素细胞中GPNMB和PMEL的表达

【摘要】:非转移性黑色素瘤糖蛋白b(glycoprotein non-metastatic melanoma protein b,GPNMB)及其同系物色素细胞特异性黑色素细胞蛋白(pigment cell-specific melanocyte protein,PMEL)对黑素体的形成和黑色素的生成具有重要的作用,而GPNMB和PMEL的功能还有待深入研究。本文旨在探索GPNMB和PMEL在不同毛色绵羊皮肤表达是否存在差异,确定紫外线b(UVB)对GPNMB和PMEL是否有影响。免疫组织化学结果表明,GPNMB在不同毛色绵羊皮肤的毛基质、内外毛根鞘、毛乳头等区域均有表达。Western印迹和qRT-PCR结果显示,黑色绵羊皮肤中GPNMB和PMEL表达量,高于白色绵羊皮肤。黑色素细胞经UVB照射后,GPNMB和PMEL的表达增高。UVB照射剂量为100 m J/cm2时,黑色素细胞活性最高;单次UVB辐射后,GPNMB和PMEL mRNA表达量在72h达到顶峰,两者的曲线变化趋势大致相同。UVB连续辐射后,GPNMB mRNA表达量在4 d达到顶峰,而PMEL mRNA表达量虽然在3 d升高,但且对GPNMB作用更明显。上述结果提示,GPNMB和PMEL在不同毛色绵羊皮肤存在差异性表达。UVB单次或者连续照射,均促进GPNMB和PMEL表达,且对GPNMB影响更显著。GPNMB和PMEL可能通过影响黑素体的成熟进而调节黑色素的生成。
[Abstract]:The non-metastatic melanoma glycoprotein (b (glycoprotein non-metastatic melanoma protein) and its congener pigment cell-specific melanocyte protein (pigment cell-specific melanocyte) play an important role in the formation of melanoma and the formation of melanin, but the functions of GPNMB and PMEL need to be further studied. The purpose of this study was to explore the difference of GPNMB and PMEL expression in sheep skin with different wool colors and to determine whether UV b (UVB) had an effect on GPNMB and PMEL. The results of immunohistochemistry showed that the expression of GPNMB and PMEL in sheep skin was higher than that in white sheep skin. Western blot and qRT-PCR showed that GPNMB and PMEL were expressed in hair matrix, inner and outer hair root sheath, hair papilla and other areas of sheep skin with different colors of wool. The results of immunohistochemistry showed that the expression of GPNMB and PMEL in black sheep skin was higher than that in white sheep skin. The expression of GPNMB and PMEL in melanocytes increased after UVB irradiation. When the dose of UVB was 100m J/cm2, the activity of melanocytes was the highest, and the expression of GPNMB and PMEL mRNA reached its peak at 72 h after single UVB irradiation. The change trend of the two curves was approximately the same. The expression of GPNMB mRNA reached its peak at 4 days after continuous radiation of UVB, while the expression of PMEL mRNA increased at 3 days, but the effect on GPNMB was more obvious. These results suggest that there are different expressions of GPNMB and PMEL in sheep skin with different color. UVB can promote the expression of GPNMB and PMEL in sheep skin by single or continuous irradiation. The effect on GPNMB is more significant. GPNMB and PMEL may regulate melanin production by affecting the maturation of melanoma.
