猪内服和静脉给予多西环素的药代动力学研究

【摘要】:以10 mg/kg体重的剂量研究猪内服盐酸多西环素饮水剂的药动学特征,采用超高效液相色谱-串联质谱法,测定猪内服与静脉给予多西环素的血浆浓度,Winnolin软件进行分析。猪单次内服多西环素饮水剂型水溶液后,MRT_(last)为(12.82±3.34)h,T1/2为(11.65±4.80)h,T_(max)为(4.13±1.89)h,Cmax为(0.87±0.48)μg/m L,AUC_(last)为(11.07±4.19)μg·h·mL~(-1)。静脉注射10 mg/kg体重多西环素水溶液后,AUClast为(33.34±8.03)μg·h·mL~(-1),T1/2为(5.68±1.32)h,MRT_(last)为(7.98±0.56)h。结果表明多西环素饮水剂水溶液的绝对生物利用度为33.2%。猪内服多西环素饮水剂后,具有体内吸收速度较快,达峰时间短,消除较慢,血中平均滞留时间较长等特点。
[Abstract]:The pharmacokinetic characteristics of doxycycline hydrochloride drinking water in pigs were studied at the dose of 10 mg/kg body weight. The plasma concentration of doxycycline hydrochloride in pigs was determined by ultra-high performance liquid chromatography-tandem mass spectrometry (HPLC-MS). The software Winnolin was used to analyze the pharmacokinetics of doxycycline hydrochloride. The MRT _ (last) was (12.82 卤3.34) h ~ (-1) and (11.65 卤4.80) h ~ (-1) (max) was (0.87 卤0.48) 渭 g / m ~ (-1) 渭 g / L AUC _ (last), (11.07 卤4.19) 渭 g / h mL~ (-1). AUC last was (33.34 卤8.03) 渭 g / h mL~ (-1) T _ 1 / 2 after intravenous injection of doxycycline solution for 10 mg/kg (5.68 卤1.32) h MRT _ (last) was (7.98 卤0.56) h. The results showed that the absolute bioavailability of doxycycline water solution was 33.2. After pigs were given doxycycline drinking water, the absorption rate was faster, the peak time was short, the elimination was slower, and the average retention time in blood was longer.
