丁香叶总酚酸提取工艺的优化及抗氧化活性测定

【摘要】:丁香叶主要来源于木樨科丁香属植物紫丁香(Syringa oblate Lindl)、朝鲜丁香(Syringa diatata Nakai)和洋丁香(Syringa vulgaris L)的干燥叶。丁香叶具有良好的抗菌消炎、抗氧化、抗病毒、保肝利胆、增强机体体液免疫、降温降压和镇咳祛痰等作用,是一种理想的广谱抗菌药,其药效已得到肯定。丁香叶胶囊剂和片剂已收录于《中华人民共和国卫生部药品标准》中药成方制剂,总酚酸作为发挥作用的药效基础,由于具有较强的抗氧化活性,已在食品领域中广泛应用。但丁香叶中总酚酸含量较低,国内外目前均无关于丁香叶总酚酸富集工艺的研究报道,因而限制了丁香叶开发和利用。因此,本实验对丁香叶提取、富集工艺进行优化,并验证总酚酸富集后抗氧化活性得到了显著提高。为丁香叶的工业化生产及作为新型的保健品和食品添加剂的开发使用提供理论基础。本试验首先采用Box Behnken响应面法对丁香叶总酚酸的乙醇提取工艺进行优化。以总酚酸提取率为考察指标,在单因素实验基础上,采用响应面分析法对乙醇浓度、提取温度、提取时间和液料比等提取条件进行了优化。结果表明,乙醇回流提取总酚酸的最佳工艺条件为:乙醇浓度44%、提取温度90℃、提取时间96min、液料比25.2:1(m L/g)、提取次数为2次,在此条件下提取率为1.64±0.23mg/g,与预测值相近。在提取工艺优化的基础上,本试验采用大孔树脂法对丁香叶总酚酸的富集工艺进行了优化。选取了D-101、X-5、HPD100、HPD 300、AB-8、S-8、NKA-9、HPD826、HPD130、HPD400和HPD600十一种树脂考察吸附特性。静态吸附试验表明S-8树脂表现出了良好的吸附、解吸能力和快速的吸附能力。用动态吸附、解吸试验优化富集工艺,优化的吸附条件:总酚酸初始浓度为2.06mg/m L;p H5.0;上样量4.3BV;上样速率2BV/h;优化的解吸参数为:去离子水、5BV、50%乙醇,流速2BV/h。按照优化的条件用S-8树脂处理过的丁香叶总酚酸干燥提取物中含量由2.75%上升到了9.18%,回收率为81.61%。试验结果表明S-8树脂对丁香叶总酚酸具有很好的富集能力,建立的方法稳定、可靠,为规模化生产提供了基础。使用DPPH·清除率、羟自由基清除率、超氧阴离子自由基清除率、Fe还原能力为考察指标来测定丁香叶总酚酸纯化前后的抗氧化活性。结果表明丁香叶总酚酸浓度越高,抗氧化活性越好,并且相同质量浓度的提取物,纯化后的抗氧化活性显著高于纯化前的活性,丁香叶具有较强的抗氧化能力。因此丁香叶可作为一种新型天然抗氧化剂。本研究首次采用响应面法优化了丁香叶总酚酸的提取工艺,并用大孔吸附树脂法优化了总酚酸的富集工艺,并测定了其抗氧化活性。为丁香叶总酚酸未来的产业化应用奠定理论基础。
[Abstract]:The clove leaves are mainly derived from the dried leaves of (Syringa oblate Lindl), (Syringa diatata Nakai) and (Syringa vulgaris L). Clove leaf is an ideal broad-spectrum antimicrobial drug with good anti-inflammatory, anti-oxidation, anti-virus, anti-liver and gallbladder, enhanced humoral immunity, lower temperature and lower blood pressure, and antitussive and expectorant effect. Clove leaf capsules and tablets have been included in the traditional Chinese medicine prescription of the Ministry of Health of the people's Republic of China. The total phenolic acid has been widely used in the field of food because of its strong antioxidant activity. The content of total phenolic acid in the leaves of Dante is relatively low, which is not related to the enrichment process of total phenolic acid in clove leaves at home and abroad, which limits the development and utilization of clove leaves. Therefore, the extraction and enrichment process of clove leaves was optimized, and the antioxidant activity of total phenolic acid was significantly improved. It provides a theoretical basis for the industrial production of clove leaves and the development and application of new health products and food additives. In this experiment, Box Behnken response surface method was used to optimize the ethanol extraction process of total phenolic acid in clove leaves. On the basis of single factor experiments, the extraction conditions of ethanol concentration, extraction temperature, extraction time and the ratio of liquid to material were optimized by using the total phenolic acid extraction rate as the index.
