不同无机磷水平和植酸酶对断奶仔猪骨骼钙磷代谢及肠道相关基因表达的影响

【摘要】:本文旨在研究不同无机磷水平和植酸酶对断奶仔猪骨骼钙磷代谢以及肠道相关基因表达的影响。试验选用224头初重(9.12±0.29 kg)相近、健康状况良好的“杜×长×大”三元杂交断奶仔猪,随机分为7个处理,每个处理4个重复,每个重复8头。处理1为对照组,饲喂基础饲粮;处理2~6保持钙水平(7.00 g/kg)不变,添加750 FTU/kg的植酸酶,无机磷添加量分别为对照组的100%、75%、50%、25%、0%(钙磷比为1.17~1.79:1);处理7为负对照,无机磷添加量为0%,且不添加植酸酶。试验期35 d。结果表明:当饲粮无机磷添加量为对照组的0%时,添加植酸酶使胫骨及肋骨磷含量极显著升高(P0.01);血清CT、OPN显著降低(P0.05)。饲粮无机磷添加量为100%条件下,添加植酸酶,骨合成生化指标BAP浓度显著升高(P0.05);骨吸收生化指标CTX-Ⅰ、OPN、TRACP-5b显著降低(P0.01)。加植酸酶的条件下(750 FTU/kg),降低无机磷的添加量,骨骼鲜重、脱脂重、骨骼钙、磷及灰分含量呈线性及二次降低(P0.05);血清CT、l,25-(OH)2D3及骨吸收生化指标CTX-Ⅰ、OPN、TRACP-5b、显著升高(P0.05),且呈线性和二次相关(P0.01);血清FGF23及骨合成生化指标BAP、BGP、OPG显著降低(P0.05),且呈线性和二次相关(P0.01)。说明低磷饲粮会减弱骨合成作用,促进骨吸收。骨合成生化指标BAP、OPG、BGP与血清P、FGF23以及胫骨钙、磷、灰分含量正相关,与血CT、l,25-(OH)2D3含量负相关;骨吸收生化指标CTX-Ⅰ、OPN、TRACP5b与血CT、l,25-(OH)2D3呈正相关,与血清P、FGF23以及胫骨钙、磷、灰分含量负相关;且BAP、OPG、BGP与CTX-Ⅰ、OPN、TRACP5b呈负相关。说明骨代谢生化指标能反应机体骨骼钙磷代谢状况。饲粮无机磷添加量为100%和0%条件下,添加植酸酶,断奶仔猪小肠NaPi-IIb mRNA的表达量显著降低(P0.01)。加植酸酶条件下,随饲粮无机磷添加量的降低,断奶仔猪小肠NaPi-IIb mRNA(P0.01)以及十二指肠、肾脏CaBP-D9k mRNA表达量(P0.05)显著升高;空肠与回肠的CaBP-D9k mRNA的表达量差异不显著(P0.05)。无机磷水平和植酸酶对断奶仔猪小肠SGLT-1 mRNA的表达量无显著影响(P0.05)。组化结果显示,NaPi-IIb蛋白主要分布于小肠吸收细胞细胞质,且沿纹状缘浓度较大,固有层部分细胞细胞质中也存在。100%、50%、0%三个磷水平,小肠绒毛吸收细胞细胞质着色逐渐变深,说明细胞内NaPi-IIb蛋白浓度随磷水平降低增大。低磷饲粮可以刺激肠道磷的主动吸收,提高其吸收效率。综上所述:植酸酶能改善断奶仔猪的骨骼性能,且在低磷条件下效果更明显;加植酸酶饲粮,降低无机磷的添加量会降低断奶仔猪骨骼钙、磷及灰分含量,并促进小肠NaPi-IIb蛋白及其mRNA的表达;骨代谢生化指标能准确反映骨代谢状况,且比常规指标更敏感。综合各项指标,在断奶仔猪饲粮中添加植酸酶可以替代25%(4.02 g/kg)的磷酸氢钙添加量。
