基于血清代谢组学研究Kisspeptin对热应激大鼠的保护作用
[Abstract]:The protective effect of neuropeptide Kisspeptin on heat stress rats was studied by metabonomics. In this experiment, the heat stress rat model was established, and the endogenous metabolites in serum were detected by gas chromatography-time-of-flight mass spectrometry (GC/TOFMS), and the control group was analyzed by principal component analysis and partial least-squares discriminant analysis. The differences of metabolites between heat stress group and Kisspeptin intervention group were used to screen out potential biomarkers. The results of metabonomics showed that the serum metabolic spectrum of heat stress group was obviously different from that of normal control group, but the serum metabolic spectrum of Kisspeptin group tended to be normal. The results of biochemical metabolic pathway analysis showed that heat stress induced abnormal metabolism of tricarboxylic acid, glucose, amino acid and nucleotide in rats. However, Kisspeptin treatment after heat stress alleviated the disorder of these metabolic pathways and tended to be normal. In addition, the results of oxidative stress index showed that Kisspeptin treatment could decrease the content of malondialdehyde (MDA) in serum and increase the activities of superoxidase dismutase (T-SOD), total antioxidant capacity (T-AOC) and glutathione peroxidase (GSH-Px). The results showed that Kisspeptin alleviated oxidative stress induced by heat stress to some extent. In this study, the protective effects of Kisspeptin on heat stress rats were studied from the point of view of metabolism and antioxidation, which provided experimental basis for healthy breeding of animals and development of safe biological products resistant to heat stress.
