抗生素所致菌群失调腹泻小鼠肠道乳糖酶活性的研究

【摘要】:为探讨抗生素造模对菌群失调小鼠肠道乳糖酶活性的影响,本研究采用硫酸庆大霉素和头孢拉定混合抗生素制备菌群失调腹泻小鼠模型,造模成功后分别提取模型组和正常组小鼠第1 d至第7 d肠道内容物及肠粘膜。将肠粘膜平均分成前、中、后3段,每段约10 cm,用ONPG法分别测定肠内容物及各段肠黏膜所含乳糖酶活性,结果表明,造模第2 d,模型组肠内容物中乳糖酶活性均明显低于正常组,第4 d开始两组差异显著(p0.05)。模型组肠前段黏膜乳糖酶活性自第3 d开始与正常组对比差异极显著(p0.01)、肠中段、肠后段黏膜乳糖酶活性分别自第2 d、第3 d开始与正常组对比差异显著(p0.05),且各肠段粘膜自第5 d开始与正常组对比差异极显著(p0.01)。上述结果表明,抗生素造模显著影响小鼠肠道乳糖酶活性,且抗生素所致菌群失调性腹泻与肠道乳糖酶活性降低有关。
[Abstract]:In order to investigate the effect of antibiotic modeling on lactase activity in intestinal tract of mice with dysbacteriosis, the model of diarrhea was established by using mixed antibiotics of gentamicin sulfate and cefradine. The intestinal contents and intestinal mucosa were extracted from the model group and the normal group from day 1 to day 7. The activity of lactase in intestinal contents and intestinal mucosa was measured by ONPG method for 10 cm, the results showed that the activity of lactase in intestinal mucosa was determined at the 2nd day of model making, and the results showed that the activity of lactase in intestinal mucosa was determined by ONPG method. The lactase activity in the intestinal contents of the model group was significantly lower than that in the normal group, and there was a significant difference between the two groups at the 4th day (p0.05). The activity of lactase in the anterior mucosa of the model group was significantly different from that in the normal group from the 3rd day (p0.01), and the activity of lactase in the middle and posterior segments of the intestine was from the 2nd day, respectively. There was a significant difference between the third day and the normal group (p0.05), and the significant difference between the intestinal mucosa and the normal group from the 5th day (p0.01). The results indicated that antibiotic modeling significantly affected the intestinal lactase activity in mice, and the decrease of intestinal lactase activity was associated with the decrease of intestinal lactase activity in patients with dysbacteriosis induced by antibiotics.
