九里明超微粉对仔猪生长性能的影响及其机理初探

【摘要】:本文以九里明为材料,超微粉碎后进行粒度检测、扫描电镜形态观察和总黄酮含量的测定。然后将九里明超微粉添加到仔猪饲料中,研究对仔猪生长性能的影响。选取体重为10 kg左右的杜×长×大三元杂交仔猪90头,按随机区组设计、公母各半的原则,将仔猪分成5个处理组,每个处理3个重复,每个重复6头仔猪。Ⅰ组饲喂基础日粮,Ⅱ组饲喂添加75 mg/kg金霉素和20 mg/kg硫酸粘杆菌素的日粮,Ⅲ、Ⅳ、Ⅴ组分别饲喂添加0.4%、0.8%、1.2%九里明超微粉的日粮。进行为期30 d的试验,研究九里明超微粉对仔猪生长性能、腹泻频率、血清生化指标、粪便微生物、营养物质消化率以及IGF-1 mRNA表达量的影响。结果如下:1.九里明超微粉的中心粒径为8.88μm,90%的超微粉粒径小于等于23.37μm,符合超微粉碎要求。扫描电镜观察发现,超微粉碎处理使九里明植物组织细胞结构破损显著,粉体粒度更小,更均匀。2.九里明超微粉总黄酮方法学试验的相对偏差值良好,适合用此方法检测九里明超微粉总黄酮。金丝桃苷的标准曲线为y=0.0534x-0.051,在3.2~16μg/mL浓度范围内有良好的线性关系(R2=0.9975)。试验发现,九里明超微粉总黄酮含量为2.72%。3.添加九里明超微粉能提高仔猪平均日增重和平均日采食量,降低料肉比,添加0.8%的九里明超微粉能显著提高仔猪的平均日增重、显著降低料肉比,且与抗生素添加组的效果相当;添加九里明超微粉降低了仔猪的腹泻频率,并且0.8%、1.2%的添加量和添加抗生素降低仔猪腹泻频率能达到显著水平;添加九里明超微粉能在一定程度上提高血清GLU、TG、T3、T4、IGF-I含量,降低血清BUN含量,其中添加0.8%的九里明超微粉能显著提高血清T3、IGF-I的含量,显著降低BUN含量。4.抗生素的添加显著降低了仔猪粪便中大肠埃希菌数量,九里明超微粉的添加也不同程度地降低了仔猪粪便中大肠埃希菌数量,试验30 d时0.8%、1.2%九里明超微粉的添加降低大肠埃希菌数达到了显著水平;与对照组相比,各处理组粪便乳酸菌数量均有所增加,但差异不显著;添加不同量的九里明超微粉不同程度地提高了仔猪各营养物质的表观消化率,添加九里明超微粉的各组间相比,添加0.8%的九里明超微粉的效果最好。5.随着九里明超微粉添加量的增加,仔猪血液IGF-I mRNA相对表达量也呈增加趋势;添加九里明超微粉的各试验组血液IGF-I mRNA相对表达量要高于空白对照组(Ⅰ组),分别提高了18.97%、24.90%、49.80%;与抗生素添加组(Ⅱ组)相比,九里明超微粉添加组血液IGF-I mRNA相对表达量也有所升高,但未达到显著差异的水平。总之,仔猪日粮中添加九里明超微粉能降低仔猪的腹泻频率,调节了仔猪肠道菌群结构,改善了仔猪肠道环境,增强了仔猪的消化吸收能力,并调节IGF-I mRNA的表达,改善了血液生化指标,促进了仔猪的生长性能。结合九里明超微粉的使用量而言,0.8%的添加量效果最好。九里明超微粉促进仔猪生长的可能机理是:九里明的药效成分黄酮等化合物抑制了肠道有害菌的增殖,促进了有益菌的生长,改善了肠道微生物环境,进而降低仔猪腹泻频率、提高了肠道对营养物质的消化吸收能力;九里明超微粉作为营养素成分作用于机体,调节IGF-I mRNA的表达,调节T3、T4和IGF-I的分泌,影响了机体对糖类、脂肪和蛋白质的代谢。在九里明超微粉药效和营养的双重作用下,仔猪的生长性能得到了促进。
