基于cox1基因序列分析山羊褐黄血蜱种系发育关系

【摘要】：为分析山羊褐黄血蜱分离株的线粒体(mt DNA)部分序列(pcox1)的遗传变异情况,并利用cox1序列分析山羊褐黄血蜱与其它蜱的种群遗传关系,本研究利用PCR扩增山羊褐黄血蜱的部分cox1片段,并对其进行测序分析。结果显示16株山羊褐黄血蜱分离株的cox1序列长度均为800 bp左右,且分离株与基因库中褐黄血蜱位于同一分支,与其它蜱类得到了很好的鉴别。本研究表明pcox1序列种内相对保守,种间差异较大,可作为山羊褐黄血蜱的种间遗传变异研究的分子遗传标记。
[Abstract]:In order to analyze the genetic variation of mitochondrial (mt DNA) partial sequence (pcox1) of Haemaphysalis nigra isolates, and to analyze the population genetic relationship between Haemaphysalis nigra and other ticks by cox1 sequence. In this study, PCR was used to amplify some cox1 fragments of Haemaphysalis bovis and to sequence them. The results showed that the cox1 sequence length of 16 isolates of Haemaphysalis goatens was about 800 bp, and the isolates were located in the same branch as Haemaphysalis bovis in the gene bank, and were well identified with other ticks. The results showed that the pcox1 sequence was relatively conserved within species and varied greatly among species, which could be used as a molecular genetic marker for the study of interspecific genetic variation of Haemaphysalis bovis.
