基于马立克病毒的传染性喉气管炎病毒载体疫苗研制

【摘要】：以表达EGFP报告基因的重组I型马立克病毒疫苗株(CVI988/Rispens)rMDV-EGFP为母本病毒,通过同源重组的方法用表达传染性喉气管炎病毒(ILTV)gB和gD基因的表达盒替换病毒rMDV-EGFP中的EGFP报告基因表达盒,分别获得了表达gB和gD基因的重组马立克病毒rMDV-ILTgB和rMDV-ILTgD。经PCR和间接免疫荧光试验鉴定,证明重组病毒中插入的目的基因大小正确,且能够表达gB和gD目的蛋白。免疫效力试验结果表明:与弱毒疫苗相比,重组病毒rMDV-ILTgB和rMDV-ILTgD均可对ILTV强毒攻击提供更好的免疫保护。重组病毒免疫组试验鸡不仅在发病率、致死率和攻毒后体重增长等指标方面与攻毒对照组均存在显著差异,而且与常规弱毒疫苗免疫组在致死率和攻毒后体重增长指标方面也存在显著差异。
[Abstract]:The recombinant Marek virus type I vaccine strain (CVI988/Rispens) rMDV-EGFP expressing EGFP reporter gene was used as female strain. By homologous recombination, the EGFP reporter gene expression cassette in the viral rMDV-EGFP was replaced by the expression cassette expressing the (ILTV) gB and gD genes of infectious laryngotracheitis virus. Recombinant Marek virus rMDV-ILTgB and rMDV-ILTgD. expressing gB and gD genes were obtained, respectively. By PCR and indirect immunofluorescence assay, it was proved that the target gene inserted in the recombinant virus was correct in size and could express gB and gD target protein. The results of immunological efficacy test showed that the recombinant virus rMDV-ILTgB and rMDV-ILTgD could provide better immune protection against ILTV attack than the attenuated vaccine. Compared with the control group, there were significant differences in the incidence, fatality rate and body weight gain between the chickens immunized with recombinant virus and those in the control group. There were also significant differences in mortality and body weight gain between vaccinated group and routine attenuated vaccine.
