板栗总苞多酚提高肉鸡抗热应激能力作用机制研究

【摘要】:背景:热应激是诱导动物机体产生氧化应激的重要因素之一,多酚的特殊结构使其具有较强的抗氧化性和清除自由基能力。本实验室的前期生化研究发现板栗总苞多酚(PICB)具有很强的清除自由基和抗氧化能力,在高温热应激情况下PICB上调AA肉鸡空肠粘膜促生长、抗氧化相关基因mRNA表达。目的:分别从体外细胞试验和体内试验两个方面研究板栗总苞多酚对IEC-6细胞的促生长、抗氧化作用和对热应激艾拔益加(AA)肉鸡血清及空肠黏膜代谢组学的影响。阐明板栗总苞多酚的促生长、抗氧化作用机制,为板栗总苞多酚作为天然植物饲料添加剂的应用奠定理论基础。方法:1、利用MTT法检测不同浓度板栗总苞多酚对IEC-6细胞增殖活力的影响并确定最佳添加量:2、利用荧光定量PCR和Western Blottin g蛋白免疫印迹技术检测板栗总苞多酚对IEC-6细胞(Ⅰ为常温对照组、Ⅱ为常温板栗总苞多酚组、Ⅲ高温对照组、Ⅳ为高温板栗总苞多酚组)抗氧化、促生长相关因子nRNA转录和蛋白表达;3、利用气相色谱-飞行时间质谱法分析研究板栗总苞多酚对热应激AA肉鸡空肠黏膜和血清中小分子代谢物差异变化的影响。结果:1、常温情况下各浓度板栗总苞多酚作用于IEC-6细胞,不同浓度细胞增殖活力与对照组相比无显著差异;高温情况下对照组显著低于多酚组(P0.05),且在浓度为180μg/ml时,多酚组细胞OD值达到最大(P0.05)并与常温对照组差异不显著。2、常温条件下,Ⅰ组HSP70和EGF mRNA表达量与Ⅱ组相比较无显著变化(P0.05);Ⅱ组SOD、EGFR和ODC mRNA表达量显著高于Ⅰ组(P0.05);Ⅱ组SOD蛋白表达量显著高于Ⅰ组(P0.05),Ⅰ组和Ⅱ组EGFR和ODC蛋白表达量基本相同(P0.05);高温热应激状态下,Ⅳ组HSP70、ODC、EGF、EGFR、SOD mRNA表达量均显著高于Ⅲ组(P0.05);Ⅳ组SOD、ODC和EGFR蛋白表达量均高于Ⅲ组。3、与对照组相比0.3%板栗总苞多酚组显著降低了AA肉鸡血清中谷氨酸含量(P0.05),并显著降低空肠黏膜中肌醇和苏糖醇含量(P0.05),同时显著提高了鸟氨酸、丝氨酸和软脂酸含量(P0.05)。结论:板栗总苞多酚能够显著提高热应激IEC-6细胞的生长性能和抗氧化性能,缓解高温对IEC-6细胞造成的损伤;饲料中添加0.3%板栗总苞多酚通过影响AA肉鸡的谷胱甘肽代谢、碳水化合物代谢、脂肪代谢以及蛋白质代谢途径,提高AA肉鸡的抗氧化性能和促生长作用,缓解高温对AA肉鸡造成危害。

The special structure of polyphenols makes it have strong antioxidant and scavenging ability of free radicals. It was found that Chestnut involucre polyphenol (PICB) had strong free radical scavenging and antioxidant activity. Under high temperature heat stress, PICB up-regulated the growth of jejunal mucosa and the expression of antioxidant related gene mRNA in the jejunum of AA broilers. Objective: to study the effects of Chestnut involucre polyphenols on the growth of IEC-6 cells in vitro and in vivo, and on the metabolism of serum and jejunum mucosa of (AA) broilers under heat stress. The mechanism of promoting growth and antioxidation of Chestnut involucre polyphenols was expounded, which laid a theoretical foundation for the application of Chestnut involucre polyphenols as natural plant feed additives.
