我国蛋用型地方鸡品种遗传多样性分析

【摘要】：对仙居鸡和白耳黄鸡2个蛋用型地方品种mtDNA D-loop区全序列进行PCR扩增和测序,分析其线粒体D-loop区多态性,并结合GenBank中洛岛红和白来航的D-loop区序列进行进化分析。结果表明:2个蛋用型地方品种mtDNA D-loop区全长1 231~1 232bp,共发现18处单核苷酸变异位点和8种单倍型,核苷酸多样性为0.006 51±0.001 09,单倍型多态性为0.781±0.011。系统发育分析发现,2个蛋用型地方品种主要分布在A、B和C 3个进化分支,B和C为优势分支,而2个标准品种则分布在A和E 2个进化分支,E为优势分支。结果提示2个蛋用型地方品种遗传多态性呈中等,与标准品种亲缘关系较远,母系来源也有所不同。这为2个品种的选育利用和有效保种提供遗传背景资料和理论依据。
[Abstract]:The whole mtDNA D-loop region of Xianju chicken and white ear yellow chicken were amplified by PCR and sequenced, the polymorphism of mitochondrial D-loop region was analyzed, and the sequence of D-loop region of Luodaohong and Bailaihang in GenBank was analyzed. The results showed that the total length of mtDNA D-loop region was 1231 BP, 18 single nucleotide mutation sites and 8 haplotypes were found in 2 native egg varieties. The nucleotide diversity was 0.006 51 卤0.001 09, and the haplotype polymorphism was 0.781 卤0.011. Phylogenetic analysis showed that two native varieties of egg type were mainly distributed in three evolutionary branches of Agna B and C, B and C were dominant branches, while two standard varieties were distributed in two evolutionary branches of A and E, and E was the dominant branch. The results suggested that the genetic polymorphisms of the two native varieties were moderate, and the genetic relationship with the standard varieties was far away, and the matriarchal origin was also different. This provides genetic background data and theoretical basis for breeding, utilization and effective conservation of two varieties.
