茶多酚对猪精液常温保存效果的影响

【摘要】:【目的】研究茶多酚(TPP)对猪精液常温保存效果的影响。【方法】手握法采集20月龄健康杜洛克种公猪精液,在基础稀释液中添加不同质量浓度的TPP(0(CK),0.01,0.02,0.04,0.08,0.16g/L)作为稀释液进行常温(17℃)保存,每隔24h检查精子活率、质膜完整率和顶体完整率,分别在精液保存0,2,4,6d时检测精液中的细菌含量,并在3d和6d时检测精液中的丙二醛含量(MDA)和总抗氧化(T-AOC)活性。【结果】添加0.02,0.04,0.08g/L TPP可明显提高猪精子活率、质膜完整率、顶体完整率和T-AOC活性,降低精液的MDA含量和细菌含量。且以0.04g/L TPP效果最佳,精液有效保存时间为6d。6d时0.04g/L TPP组猪精子活率、质膜完整率、顶体完整率、丙二醛(MDA)含量、总抗氧化(T-AOC)活性和细菌含量分别为60.02%,46.40%,75.25%,2.39nmol/L,2.31U/mL和2.19×106 CFU/mL。【结论】茶多酚能有效提高猪精液常温保存效果,最适添加量为0.04g/L。
[Abstract]:[objective] to study the effect of tea polyphenols (TPP) on the preservation of porcine semen at room temperature. [methods] 20 month-old healthy Duroc boar semen was collected by hand grip method, and TPP (0 (CK),) was added to the basic diluent. The sperm motility rate, plasma membrane integrity rate and acrosome integrity rate were examined every 24 hours, and the bacterial content in semen was measured after 6 days of semen preservation, respectively, at 0.16 g / L 0.08 ~ 0.08 ~ 0.16 g / L as diluent solution, and the sperm motility rate, plasma membrane integrity rate and acrosome integrity rate were examined every 24 hours. The content of malondialdehyde (MDA) and the activity of total antioxidant (T-AOC) in semen were measured at 3 and 6 days. [results] the sperm motility and plasma membrane integrity were significantly increased by adding 0.02n0.04 + 0.08g / L TPP. Acrosome integrity rate and T-AOC activity decreased MDA content and bacterial content in semen. The best effect was 0.04g/L TPP, and the sperm motility, plasma membrane integrity, acrosome integrity, malondialdehyde (MDA) (MDA) content in 0.04g/L TPP group were the best when the effective preservation time of semen was 6d.6d.
