恩拉霉素高效液相色谱(HPLC)检测方法的建立

【摘要】:恩拉霉素是由放线菌(Streptomyces fungicidicus)发酵而得的一类多肽类抗生素,由于其独特的分子结构,对革兰氏阳性菌有很强的活性,特别是对肠道内的有害梭菌有很强的抑杀作用。作为全新的饲料添加剂,恩拉霉素能促进猪、鸡等的生长,广泛用于畜牧业。目前恩拉霉素的含量测定是按照农业部1999年颁发的《进口兽药质量标准》方法进行,本研究根据恩拉霉素的结构特点,通过实验,建立了恩拉霉素的高效液相色谱检测法。筛选优化色谱条件为:色谱柱:Thermo C18,5μm,4.6mm×250mm;流动相:乙腈:0.05mol/L磷酸二氢钠溶液=30:70;紫外检测波长267nm;柱温:30℃;流速:1.0mL/min;进样量:20μL。在该条件下,检测结果显示恩拉霉素A和恩拉霉素B在12.5~200μg/mL范围内线性关系良好,恩拉霉素A和恩拉霉素B的回归方程分别为A1=11587x+18926(R2=0.998)和A2=35969x-46745(R2=0.996);恩拉霉素A的平均回收率为98.9%,恩拉霉素B的平均回收率为98.7%;相对标准偏差(RSD)为0.27%。实验结果证明采用建立的高效液相色谱法(HPLC)检测恩拉霉素含量具有线性范围宽、分析时间短、样品前处理简便、定量结果准确、重复性好、稳定性好和回收率高等特点,可以作为恩拉霉素含量检测的质控方法。同时将高效液相色谱法与现行标准微生物检测法进行了比较,进一步证明高效液相色谱法(HPLC)操作简便、快速、精准的特点,为其质量控制提供了更为可靠的依据。
[Abstract]:Enramycin is a kind of polypeptide antibiotics fermented by actinomycetes (Streptomyces fungicidicus). Because of its unique molecular structure, it has strong activity against Gram-positive bacteria, especially against harmful Clostridium perfringens in intestinal tract. As a new feed additive, Enramycin can promote the growth of pigs, chickens and so on, and is widely used in animal husbandry. At present, the content of Enramycin was determined according to the quality Standard of imported Veterinary drugs issued by the Ministry of Agriculture in 1999. according to the structural characteristics of Enramycin, a high performance liquid chromatography (HPLC) method for the determination of Enramycin was established.
