Hemin对妊娠期高血压疾病大鼠HO-1的表达、血压及24h尿蛋白的影响

【摘要】:目的:(1)探讨血红素氧合酶-1与大鼠妊娠期高血压疾病的相关性;(2)探讨正铁血红素(Hemin)对妊娠期高血压大鼠血红素氧合酶.1(HO-1)的表达、血压及24h尿蛋白的影响。 方法:(1)建模及干预:正常SD雌性大鼠30只与雄性大鼠1:1合笼,雌鼠阴道分泌物发现精子计为妊娠第0天,妊娠第12天测量孕鼠血压并收集24h尿量。妊娠第14天将孕鼠随机分为三组:L-NAME组(A)、L-NAME+Heminta (B)、正常妊娠组(C),A、B组予亚硝基左旋精氨酸甲酯(L-NAME)80mg/kg/d灌胃建立大鼠妊娠期高血压模型,C组予等量生理盐水灌胃。妊娠第16天再次测量各组孕鼠血压,A、B组血压升高达到高血压诊断标准者纳入实验,继续每日灌胃L-NAME80mg/kg/d,共7天,B组同时于妊娠第16天始给予Hemin30mg/kg腹腔注射4天。(2)标本采集及指标检测:妊娠第12天、妊娠第16天、妊娠第20天测量各组孕鼠血压并收集24h尿量,用自动生化分析仪测定24h尿蛋白;妊娠第20天收集尿量完毕后取孕鼠胎盘进行HE染色观察胎盘形态学变化及免疫组织化学法检测胎盘组织中HO.1的定位和表达;腹主动脉采血用ELISA方法检测血清中HO.1的表达。 结果:(1)获妊娠大鼠21只,随机分为三组:L-NAME组(A)、 L-NAME+Hemin组(B)、正常妊娠组(C),每组7只;(2)血压(mmHg):妊娠第16天A、B组各7只孕鼠血压均升高达大鼠高血压诊断标准,均纳入实验,A、B组孕鼠血压均值均高于C组,且均较同组妊娠第12天的基础血压明显升高,差异有统计学意义(P0.05),建模成功;妊娠第20天A、B组孕鼠血压仍高于C组,B组血压较A组下降且低于同组妊娠第16天血压,差异均有统计学意义(P0.05)。(3)24h尿蛋白(mg/24h):妊娠第20天A、B组孕鼠24h尿蛋白较C组明显升高,且均较同组妊娠第12天及妊娠第16天明显升高,差异有统计学意义(P0.05);妊娠第20天B组较A组24h尿蛋白虽有下降趋势,但差异无统计学意义(P0.05)。(4)血清中HO.1的表达:A、B、C组HO-1表达量依次增多,三组两两比较差异均有统计学意义(P0.05)。妊娠第20天孕鼠24h尿蛋白与血清中HO-1表达之间存在负相关关系,Pearson相关系数为.0.603。(5)免疫组织化学染色提示HO-1主要表达于胎盘海绵滋养层细胞,A、B、C组HO-1蛋白表达依次增多,三组两两比较差异均有统计学意义(P0.05)。 结论:(1)HO-1与大鼠妊娠期高血压疾病具有相关性,可能为妊娠期高血压疾病发病机制之一;(2)Hemin能够上调HO.1的表达,降低血压,但对24h尿蛋白的影响还需进一步探讨,可能对妊娠期高血压疾病具有一定的治疗作用。
[Abstract]:Objective: (1) to explore the correlation between heme oxygenase -1 and hypertensive disorder complicating pregnancy. (2) to explore the effect of hemin (Hemin) on heme oxygenase.1 (HO-1) expression, blood pressure and 24h urinary protein in hypertensive rats.
Methods: (1) modeling and intervention: normal 30 female SD rats and male rats were mated female rats 1:1, vaginal secretions for sperm was found on day zeroth of pregnancy, Twelfth days of blood pressure measurement in pregnant rats and collect 24h urine pregnancy. Pregnancy fourteenth days pregnant rats were randomly divided into three groups: group L-NAME (A), L-NAME+Heminta (B), normal pregnancy group (C), A, B group were treated with L-NAME (L-NAME 80mg/kg/d) to establish the rat model of gastric lavage in hypertensive pregnancy, C group was treated with normal saline. The sixteenth day of pregnancy again measure blood pressure in pregnant rats of each group A, B group up to the diagnostic criteria of hypertension were included in the experiment, to gavage L-NAME80mg/kg/d, a total of 7 days, the B group on the sixteenth day of pregnancy was given intraperitoneal injection of Hemin30mg/kg for 4 days. (2) detection of specimen collection and index: Twelfth days of pregnancy, pregnancy sixteenth days, pregnancy twentieth days to measure the blood pressure of pregnant rats and collect group 24h urine volume, with Determination of 24h urine protein automatic biochemical analyzer; the expression and localization of the twentieth day of pregnancy to collect HO.1 urine volume after the placenta in pregnant rats by HE staining to observe the change of placental morphology and immunohistochemistry in the placenta tissue; the expression of abdominal aorta with ELISA method for detection of HO.1 in serum.
