【摘要】：目的：通过检测CIP2A与c-Myc在子宫内膜样腺癌中的表达情况，，分析其与临床病理参数的关系，并初步探讨两因子在子宫内膜样腺癌中发生发展的作用。方法：采用实时定量逆转录聚合酶反应方法检测80例子宫内膜样腺癌和43例正常子宫内膜中的CIP2A mRNA的表达情况，并比较它们的差异。采用免疫组织化学方法检测55例子宫内膜样腺癌（Endometrial adenocarcinoma，EAC），20例子宫内膜不典型增生（Endometrial atypical hyperplasia，EAH）和24例正常子宫内膜（Normal endometrium，NE）中CIP2A及c-Myc蛋白的表达情况，比较它们的差异；并分析CIP2A与c-Myc在子宫内膜样腺癌中的表达与临床病理参数的关系及两者的相关性。所有数据均通过SPSS17.0统计软件进行t检验、秩和检验及Spearman等级相关分析，以P0.05表示差异有统计学意义。结果： 1. CIP2AmRNA在子宫内膜样腺癌中相对表达水平明显高于正常子宫内膜组织，差异具有统计学意义（P=0.016）。 2. CIP2A及c-Myc蛋白在NE、EAH、EAC组织中表达强度呈上升趋势，差异具有统计学意义（P0.05）；在EAC组织中，CIP2A、c-Myc的表达强度均与肌层浸润深度、临床分期及组织分化程度有关（P0.05），而与淋巴结有无转移、年龄无明显相关性(P0.05)； 3. Spearman等级相关分析显示在EAC组织中CIP2A与c-Myc的表达强度呈正相关（r=0.607，P0.0001）。结论：CIP2A与c-Myc在子宫内膜样腺癌中过表达，两者呈正相关；CIP2A可能通过稳定c-Myc蛋白参与子宫内膜癌的发生和发展。
[Abstract]:Objective: to detect the expression of CIP2A and c-Myc in endometrial adenocarcinoma and to analyze the relationship between CIP2A and clinicopathologic parameters, and to explore the role of two factors in the occurrence and development of endometrial adenocarcinoma. Methods: the expression of CIP2A mRNA in 80 cases of endometrial adenocarcinoma and 43 cases of normal endometrium was detected by real-time quantitative reverse transcriptase polymerase reaction. The expression of CIP2A and c-Myc protein in 55 cases of Endometrial adenocarcinoma (EAC) and 20 cases of Endometrial atypical hyperplasia EAH (EAH) and 24 cases of normal endometrium (NE) were detected by immunohistochemical method. The relationship between the expression of CIP2A and c-Myc in endometrioid adenocarcinoma and clinicopathological parameters was analyzed. All the data were analyzed by SPSS 17.0 statistical software for t test, rank sum test and Spearman rank correlation analysis. Results: 1. The relative expression of CIP2 mRNA in endometrial adenocarcinoma was significantly higher than that in normal endometrial tissue (P0. 016). 2. The expression intensity of CIP2A and c-Myc protein increased in EAH EAC tissue, the difference was statistically significant (P0.05), the expression intensity of CIP2A and c-Myc protein were related to the depth of myometrium invasion, clinical stage and tissue differentiation (P0.05), but correlated with lymph node metastasis, and the expression intensity of CIP2A and c-Myc protein in EAC tissue was correlated with the depth of myometrial invasion, the clinical stage and the degree of tissue differentiation (P0.05). Age had no significant correlation (P0.05); 3. Spearman grade correlation analysis showed that there was a positive correlation between the expression of CIP2A and c-Myc in EAC tissues (r = 0.607, P 0.0001). Conclusion the overexpression of CIP2A and c-Myc in endometrial adenocarcinoma is positively correlated with the formation and development of endometrial carcinoma by stabilizing c-Myc protein.
