巨细胞病毒感染对妊娠妇女调节性免疫细胞的作用研究

【摘要】：目的研究巨细胞病毒(CMV)感染对妊娠患者调节性免疫细胞的作用。方法收集2015年5月-2016年9月来医院就诊妊娠患者的临床资料,抗原pp65检测阴性的30例患者为对照组;抗原pp65检测阳性的30例患者为CMV感染组,采用梯度离心方法分离患者外周血淋巴细胞,CD_4、CD_(25)和Foxp_3流式抗体标记调节性T细胞(Treg细胞);CD_(19)和TGF-β流式抗体标记调节性B细胞(Breg细胞);CD_4、CXCR_5和Foxp_3流式抗体标记滤泡调节性T细胞(Tfr细胞)。结果 CMV感染组CD_4~+CD_(25)~+Foxp_3~+的Treg细胞比例显著低于对照组,下降了31.9%,差异有统计学意义(P0.05);CMV感染组CD_(19)~+TGF-β~+的Breg细胞比例显著低于对照组,下降了78.5%,差异有统计学意义(P0.05);CMV感染组中CD_4~+CXCR_5~+Foxp_3~+的Tfr细胞比例显著低于对照组,下降了65.1%,差异有统计学意义(P0.05)。结论 CMV感染后,妊娠患者体内Treg细胞、Breg细胞和Tfr细胞等调节性免疫细胞的比例均显著下降,可能对胎儿的生长发育造成影响。
[Abstract]:Objective to study the effect of cytomegalovirus (CMV) infection on regulatory immune cells in pregnant women. Methods from May 2015 to September 2016, the clinical data of pregnant women who came to hospital from May 2015 to September 2016 were collected. CD4CXCR5 and FoxpS3 flow antibody labeled regulatory T cells (Treg cells) and TGF- 尾 labeled regulatory B cells (Breg cells) were isolated from peripheral blood lymphocytes of patients by gradient centrifugation. Results in CMV infected group, the percentage of CD4 ~ CD25 ~ 3 ~ Treg cells in CMV infected group was significantly lower than that in control group (P 0.05). The percentage of CD19 ~ TGF- 尾 ~ Breg cells in CMV infected group was significantly lower than that in control group, and the percentage of CD19 ~ TGF- 尾 ~ cells in CMV infected group was significantly lower than that in control group. The percentage of Tfr cells of CD4 ~ CXCR-5- Foxp3 ~ in CMV infected group was significantly lower than that in control group, and decreased by 65.1, the difference was statistically significant (P0.05). Conclusion after CMV infection, the percentage of regulatory immune cells such as Treg cells and Tfr cells in pregnant women decreased significantly, which may affect the growth and development of fetuses.
