不同浓度万古霉素对大鼠脂肪干细胞体外定向诱导分化为成骨细胞的影响

【摘要】: 目的:研究大鼠脂肪干细胞(ADSCs)在体外分离培养、诱导分化为成骨细胞及成脂细胞的能力,初步观察不同浓度万古霉素对其增殖、成骨分化的影响 方法:将大鼠ADSCs放入成脂诱导液中培养,采用油红O染色观察其成脂分化能力;同时将大鼠ADSCs放入不同浓度的万古霉素,以及添加了成骨诱导剂的培养液中培养,采用MTT法、不同时相点测定ALP值、Von Kossa染色法观察万古霉素对ADSCs的成骨影响。 结果:SD大鼠ADSCs可在不同浓度万古霉素溶液中生长,当万古霉素浓度≤800μg/ml时,万古霉素对ADSC增殖无影响,当浓度≥1000μg/ml时,才对ADSC的增殖有一定影响。万古霉素≤1000μg/ml时,实验组与对照组在不同时相点ALP无明显差异。通过向成脂细胞及成骨细胞定向诱导后,油红染色、碱性磷酸酶染色和vonKossa染色阳性。 结论:大鼠脂肪干细胞能向脂肪细胞和成骨细胞诱导分化;万古霉素在一定浓度范围内对ADSC增殖、成骨分化影响较小,当达到某一浓度高值时才对其产生影响,是预防和治疗骨科临床感染的较好选择。
[Abstract]:Objective: to study the ability of rat adipose stem cells (ADSCs) to differentiate into osteoblasts and adipoblasts in vitro, and to observe the effects of vancomycin at different concentrations on the proliferation and osteogenic differentiation of adipose stem cells. Methods: rat ADSCs was cultured in fat-forming medium, and the ability of adipogenic differentiation was observed by oil red O staining, and rat ADSCs was cultured in different concentrations of vancomycin and medium supplemented with osteogenic inducer. MTT method was used. The effect of vancomycin on ADSCs osteogenesis was observed by Von Kossa staining at different phase points.
