【摘要】:背景术后认知功能障碍(POCD)是术后常见的并发症之一,严重影响患者术后生活质量并增加患者病死率。年龄是已知引起POCD发生的主要因素之一,但引起POCD的具体机制仍不清楚。目的观察反复七氟烷暴露对老年大鼠学习记忆功能的影响,以及对海马区Aβ和p-tau蛋白表达的影响,探讨其对大鼠学习记忆影响的可能机制。方法20月龄SD雄性老年大鼠20只,将其随机分为七氟烷组(S组)及对照组(C组),每组10只。于20月龄时,将S组大鼠暴露于2.1%七氟烷+100%氧气中2h,对照组只给予氧气吸入,每周一次。于第四次七氟烷暴露后一周,运用Morris水迷宫系统测定大鼠空间学习记忆功能,然后,取大脑海马组织,通过免疫组化法测定海马组织CA1区Aβ和p-tau的分布和表达水平。结果与C组比较,S组大鼠逃避潜伏期无明显差异(P0.05),穿越平台次数更多但无显著差异(P0.05),平台所在象限停留时间更长(P0.05)。与C组比较,S组Aβ、p-tau蛋白和Aβ/p-tau的水平没有明显改变(P0.05)。结论反复暴露于2.1%的七氟烷可延缓老年大鼠记忆功能的衰退,对海马CA1区Aβ和p-tau蛋白的表达无明显影响,其机制有待进一步的研究。
[Abstract]:The background of postoperative cognitive dysfunction (POCD) is one of the common complications postoperatively, seriously affect the life quality of patients and increase the mortality of patients. Age is known to cause one of the main factors of POCD, but the specific mechanisms induced by POCD is still unclear. Objective To observe the effect of repeated seven halothane exposure on learning and memory function in aged rats the effect on the expression of A in hippocampus and beta and p-tau protein, and explore the possible mechanism of memory effects on rat study. Methods 20 month old male SD 20 rats were randomly divided into seven sevoflurane group (S group) and control group (C group), 10 rats in each group. At the age of 20 months, S rats were exposed to 2.1% isoflurane seven +100% oxygen 2h, the control group was given oxygen inhalation, fourth times each week. In the seven week after halothane exposure, using Morris water maze learning and memory function of rats were measured, then take the space, the hippocampus group Fabric, through the immune group distribution determination of hippocampus tissue CA1 A beta and p-tau method and the expression level. Results compared with C group, S group rats had no significant difference in escape latency (P0.05), the number of cross platform more but no significant difference (P0.05), platform quadrant longer residence time (P0.05). With the C group, S group, A beta, p-tau did not significantly alter the A protein and beta /p-tau level (P0.05). Conclusion the 2.1% recession repeated exposure to seven isoflurane can delay memory function in aged rats, had no effect on the expression of CA1 in hippocampus region of A beta and p-tau protein, the mechanism need further research.
