羟基喜树碱预防兔膝关节术后瘢痕粘连的最佳浓度

【摘要】:目的:膝关节术后,羟基喜树碱可以通过抑制成纤维细胞增殖来减少关节粘连。但是,羟基喜树碱预防膝关节粘连的最佳浓度仍然不清。故本研究拟探讨羟基喜树碱预防膝关节术后瘢痕粘连的最佳浓度。方法:将60只新西兰大白兔随机分为5组。手术去除股骨髁两侧的皮质骨,范围为10mm×10mm大小,暴露松质骨。将含有不同浓度羟基喜树碱(0.1 mg/ml,0.5 mg/ml,1.0 mg/ml,2.0 mg/ml)或生理盐水的棉片作用于手术区域10分钟。术后四周,采用大体评分、羟脯氨酸含量测定和组织学观察判断膝关节粘连程度。结果:所有动物术后恢复良好,饮食及饮水量正常,未发现有实验动物死亡、切口感染及皮肤坏死等现象。不同浓度组的羟基喜树碱均可以减少术后膝关节粘连,1.0mg/ml, 2.0mg/l羟基喜树碱组的膝关节粘连程度明显低于其他各组,且在1.0mg/ml浓度组之前随着药物浓度的不断增加,膝关节瘢痕粘连的程度越低,而1.0 mg/ml和2.0 mg/ml浓度组之间的瘢痕粘连程度没有统计学差异。结论:局部应用1.0 mg/ml羟基喜树碱或许是预防兔膝关节术后关节粘连的最佳浓
[Abstract]:Objective: after knee surgery, hydroxycamptothecin can reduce adhesion by inhibiting fibroblast proliferation. The best concentration of hydroxycamptothecin for preventing adhesion of knee joint is still unclear. Therefore, this study aims to explore the best concentration of hydroxycamptothecin in preventing scar adhesion after knee joint operation. Methods: 60 New Zealand rabbits were randomly divided into 5 groups. Removal of cortical bone on both sides of femoral condyle, The size of 10 mm 脳 10 mm exposed cancellous bone. A cotton sheet containing 0.1 mg / ml of different concentrations of HCPT (0.5 mg / ml) or normal saline was applied to the surgical area for 10 minutes. Results: all the animals recovered well, their diet and drinking water were normal, and no experimental animals were found to die. Different concentrations of hydroxycamptothecin can reduce postoperative knee joint adhesion 1.0 mg / ml, 2.0 mg / l hydroxycamptothecin group is significantly lower than other groups, the degree of knee joint adhesion is significantly lower than other groups. And before 1.0 mg / ml concentration group, with the increasing of drug concentration, the degree of knee joint scar adhesion was lower. However, there was no significant difference in the degree of scar adhesion between 1.0 mg/ml and 2.0 mg/ml groups. Conclusion: local application of 1.0 mg/ml hydroxycamptothecin may be the best concentration for preventing articular adhesion after knee joint surgery in rabbits.
