大鼠肝损伤过程中肝组织神经营养因子Neuritin水平变化及其意义

【摘要】:目的:通过SD大鼠肝损伤造模来探讨大鼠肝损伤过程中神经突蛋白(Neuritin)在肝脏组织中随着不同时限的表达,根据其含量的变化来判断神经突蛋白(Neuritin)在肝脏损伤后的意义,为深入研究肝细胞再生与肝脏系统可塑性的分子机制奠定理论基础;为肝细胞损伤后的再生修复研究提供新思路。方法:由新疆医科大学动物饲养中心提供的6周龄、清洁级的SD大鼠126只随机分三组,空白组、非肝损伤组和肝损伤组各组42SD大鼠,空白组不给任何处理,非肝损伤组在大鼠股骨处同一部位同一力度致大鼠股骨骨折,肝损伤组使用自制改良BIM-IV型生物撞击建立大鼠肝损伤模型,模型建立后分成6h,12h,1d,2d,3d,5d共6个亚组分别处死大鼠,再经HE染色、免疫组化化学染色并检测血清中丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST),观察各个时间点肝脏组织中Neuritin变化及表达水平。结果:根据造模后SD大鼠血清中ALT、AST显示:肝损伤组血清中ALT、AST 6h时开始升高,12h小时升高达到最高值,1d-2d时开始下降但仍高于空白组、非肝损伤组(P0.01),3d-5d时趋向正常水平。免疫组化示:肝损伤组neuritin蛋白6 h呈阳性表达,12 h呈强阳性,1d-3d呈弱阳性表达,但仍高于空白组与对照组(P0.01),5d时肝损伤组neuritin的表达趋向正常水平。HE染色示:空白组和非肝损伤组肝小叶结构清楚,没有明显损伤。肝损伤组肝细胞,出现明显气球样变,肝细胞大片状坏死甚至出现细胞核脱失,细胞坏死。结论:1.本实验证明在大鼠肝损伤后均引起肝组织中Neuritin的表达量增加并且与大鼠肝脏损伤程度变化有一定关系。2.通过本研究发现神经营养因子Neuritin在大鼠肝损伤细胞周围有很高的表达,和肝损伤血清中ALT、AST水平变化一致,由此推断Neuritin可能在肝细胞的再生修复或增殖过程中起着重要作用。
[Abstract]:Objective: to investigate the expression of neurite protein Neuritinin in rat liver tissue with different time limits, and to determine the significance of neurite protein Neuritin after liver injury according to the changes of its content.It provides a theoretical basis for the further study of the molecular mechanism of hepatocyte regeneration and liver system plasticity, and provides a new idea for the regeneration and repair of liver cells after injury.Methods: a total of 126 SD rats, aged 6 weeks, provided by the Animal feeding Center of Xinjiang Medical University, were randomly divided into three groups: blank group, non-hepatic injury group and liver injury group.In the non-hepatic injury group, the femur fracture was induced by the same force at the same position of the femur of the rats. The liver injury group established the rat liver injury model by using self-made modified BIM-IV biological impact. After the establishment of the model, the rats were divided into 6 subgroups, 6 subgroups were divided into 6 subgroups respectively.After HE staining and immunohistochemical staining, serum alanine aminotransferase (alt) and aspartate transaminase (AST) were detected. The changes and expression of Neuritin in liver tissue were observed at different time points.Results: according to the alt AST in the serum of SD rats after model making, the alt AST in the serum of the liver injury group began to rise at 6 h and reached the highest value at 12 h. It began to decrease but was still higher than that of the blank group at 1 d ~ 2 d. The level of alt AST in the serum of the non-hepatic injury group reached normal level at 3 d ~ 5 d.Immunohistochemistry showed that the expression of neuritin protein in liver injury group was strongly positive at 6 h and strong positive at 12 h. The expression of neuritin protein was weak at 1 d ~ 3 d after liver injury.However, the expression of neuritin in liver injury group was higher than that in blank group and control group for 5 days. The results of HE staining showed that the structure of hepatic lobules in blank group and non-hepatic injury group was clear and no obvious damage was found.In the group of liver injury, there were obvious balloon degeneration, large slice necrosis and even loss of nucleus and necrosis of the cells.Conclusion 1.This study demonstrated that the expression of Neuritin increased after liver injury in rats and was related to the degree of liver injury in rats.It was found that the expression of neurotrophic factor Neuritin was very high around the rat liver injury cells, which was consistent with the changes of alt AST level in the serum of liver injury. It was concluded that Neuritin may play an important role in the regeneration and proliferation of hepatocytes.
