环氧合酶-2选择性抑制剂塞来昔布防治大鼠创伤性异位骨化的机制

【摘要】:目的 探究环氧合酶(Cox)-2选择性抑制剂塞来昔布对大鼠创伤性异位骨化的防治机制。方法 创伤性异位骨化大鼠予塞来昔布治疗,分别采取喂食2、6、10 w的不同方案,治疗后观察各组肢体的皮温、肿胀程度及X线表现,并通过实时定量PCR及Western印迹实验,检测跟腱组织骨形态发生蛋白(BMP)-4的表达含量。结果 塞来昔布组皮温、肿胀程度、新骨形成均显著低于对照(生理盐水处理)组(P0.05)。同时,随着塞来昔布处理时间的延长,皮温均明显降低、肿胀程度及新骨形成情况均明显减轻;实时定量PCR及Western印迹检测显示:与对照组比较,塞来昔布组跟腱组织BMP-4的表达明显下调,并且随时间的延长,BMP-4的表达也相应下调。结论 塞来昔布可通过降低大鼠跟腱组织BMP-4的表达以干预异位骨化。
[Abstract]:Objective to investigate the protective mechanism of celecoxib, a selective inhibitor of cyclooxygenase-2, on traumatic ectopic ossification in rats. Methods Rats with traumatic ectopic ossification were treated with celecoxib for 10 weeks. The skin temperature, swelling degree and X ray appearance of each group were observed, and real-time quantitative PCR and Western blot were used. The expression of bone morphogenetic protein (BMP- 4) in Achilles tendon was detected. Results the skin temperature, swelling and new bone formation in celecoxib group were significantly lower than those in control group (P 0.05). At the same time, with the prolongation of celecoxib treatment time, the skin temperature was significantly decreased, the swelling degree and new bone formation were significantly reduced, and real-time quantitative PCR and Western blot analysis showed that: compared with the control group, In celecoxib group, the expression of BMP-4 in Achilles tendon was down-regulated, and the expression of BMP-4 was down-regulated with time. Conclusion Celecoxib can interfere with ectopic ossification by decreasing the expression of BMP-4 in Achilles tendon.
