AMPK-SIRT1信号通路在脊髓损伤后神经细胞自噬和细胞凋亡调节中的作用

目的:脊髓损伤(spinal cord injury,SCI)是一种严重中枢神经系统损伤,其发病率和死亡率较高,已成为一个世界性的医学难题。虽然关于此疾病的治疗研究已经在动物模型中取得了重要进展,但是目前尚无有效促进人类脊髓损伤后功能恢复的措施。脊髓继发性损伤机制的研究是开发有效治疗策略的前提,因此,SCI研究领域的首要任务是阐明SCI后复杂的病理学调控机制。目前大量研究表明5’-腺嘌呤核苷酸激活的蛋白激酶(Adenosine 5‘-monophosphate-activated protein kinase,AMPK)/沉默信息转录调节因子1(silent information regulator of transcription 1,SIRT1)信号通路参与多种疾病,例如,糖尿病,脂肪肝和肿瘤等疾病中细胞自噬的调控,但是在中枢神经损伤中关于此信号通路的报道非常少。最近,研究发现啮齿类动物SCI后AMPK和SIRT1的表达异常,但是AMPK-SIRT1通路如何调控SCI后神经细胞自噬尚不清楚。本研究首先检测SCI后体内和体外模型中AMPK和SIRT1的时间表达趋势,进一步采用AMP...
