KLD-12多肽/TGF-β1纳米纤维凝胶对纤维环细胞增殖、表型及功能影响的实验研究

目的:本实验旨在探究KLD-12多肽/TGF-β1纳米纤维凝胶在体外实验中对兔纤维环细胞增殖、表型及功能的影响,寻找修复椎间盘退变的理想种子细胞、细胞支架及生长因子。方法:胰蛋白酶消化法提取6月龄新西兰大白兔腰椎纤维环内层细胞,体外培养至第3代,与KLD-12多肽纳米纤维凝胶及TGF-β1复合,制备KLD-12多肽/TGF-β1/纤维环细胞纳米纤维凝胶(实验组)和KLD-12多肽/纤维环细胞纳米纤维凝胶(对照组),体外培养14 d。倒置显微镜观察凝胶内细胞形态变化;细胞计数试剂盒(cell count kit 8,CCK-8)法检测细胞增殖活性;钙黄绿素(Calcein-AM)/碘化丙啶(PI)双荧光染色观察凝胶内活细胞与死亡细胞,并计算活细胞比率;阿尔新蓝法检测培养基中葡萄糖胺聚糖(Glycosaminoglycan,GAG)含量;免疫荧光染色观察Ⅱ型胶原分泌情况;实时荧光定量PCR(real-time quantitative PCR,RT-q PCR)检测纤维环细胞中Ⅱ型胶原及聚蛋白多糖(Aggrecan)基因表达情况。结果:(1)倒置显微镜观察复合于凝胶中的纤维环细胞呈圆形,少...
