血液肿瘤患者WNK1-WAC易位突变分析

【摘要】：[目的]检测我科血液肿瘤患者中WNK1-WAC基因易位突变的发生率和分布情况,结合临床资料试图分析其在ANLL及其他血液肿瘤的致病及复发机理。[方法]收集2014年4月-2015年5月在昆明医科大学第一附属医院血液内科收治住院的300例不同类型血液肿瘤患者的骨髓液样品及10例非恶性血液肿瘤患者骨髓液样品(作对照)。采用PCR、RT-PCR结合琼脂糖凝胶电泳法检测血液肿瘤患者中WNK1-WAC基因易位突变并分析。[结果]300例不同血液肿瘤患者(141例AML、32例ALL、30例CML、14例CLL、18例MDS、25例MM、40例NHL)及10例非恶性血液肿瘤患者中均未检测到WNK1-WAC基因易位突变。[结论]在我科血液肿瘤患者中,未检测到WNK1-WAC基因易位突变,可能是由于样本量小,或在该人种中,无WNK1-WAC基因易位突变,疾病的发生发展与该基因无关。
[Abstract]:[objective] to detect the incidence and distribution of WNK1-WAC gene translocation mutation in patients with hematologic tumors in our department. Combined with clinical data, this paper attempts to analyze the pathogenesis and recurrence mechanism of ANLL and other blood tumors. [methods] 300 cases of different types of patients admitted to the Department of Hematology, first affiliated Hospital of Kunming Medical University from April 2014 to May 2015 were collected. Bone marrow fluid samples from patients with hematologic tumors and bone marrow fluid samples from 10 patients with non-malignant hematologic tumors were used to detect translocation mutations of WNK1-WAC gene in patients with hematologic tumors by RT-PCR and agarose gel electrophoresis. WNK1-WAC gene translocation mutations were not detected in 141 patients with different blood tumors, 30 patients with WNK1-WAC, 30 patients with CML, 14 patients with WNK1-WAC, 18 patients with MDS, 25 patients with MML and 40 patients with non-malignant hematological tumors. [conclusion] in our department, no translocation mutation of WNK1-WAC gene was detected in patients with hematologic tumors. No translocation mutation of WNK1-WAC gene was detected, which may be due to the small sample size or the absence of translocation mutation of WNK1-WAC gene in this population, and the occurrence and development of the disease have nothing to do with the gene.
