肥大细胞膜稳定剂对大鼠糖尿病心肌病变的影响

【摘要】:目的研究肥大细胞膜稳定剂酮替芬对大鼠糖尿病心肌病(DCM)的影响及可能机制。方法将45只雄性Wistar大鼠随机分为正常对照组(NC组)、DCM组和肥大细胞膜稳定剂组(Ket组)。NC组给予普通饮食,DCM组予以高脂高糖饮食加一次性腹腔注射链脲佐菌素(STZ)建立DCM大鼠模型,Ket组为DCM大鼠模型建模成功后给予0.025%的酮替芬溶液1 mg/kg腹膜腔内注射,2次/d,连续注射8 w。同时,NC组和DCM组给予等体积的生理盐水空白对照注射。实验完成后处死全部大鼠,剪下左心室,称取左心室重量计算全心指数和左心室重量指数,在电镜下对各组心肌组织进行观察分析,应用酶联免疫吸附法(ELISA)检测各组大鼠血清肿瘤坏死因子(TNF)-α、活性氧簇(ROS)表达,甲苯胺蓝改良染色法比较肥大细胞数量分布。结果 DCM组全心指数显著高于NC组和Ket组(均P0.05);DCM组左心室重量指数显著高于NC组和Ket组(均P0.05)。电镜下NC组大鼠心肌细胞核椭圆形清晰可见,闰盘连续,肌丝致密,胞内可见大量形态结构正常的线粒体;DCM组大鼠心肌细胞核固缩,线粒体肿胀,显示不清晰,闰盘扭曲断裂,肌丝溶解;Ket组大鼠心肌细胞核无明显肿胀,核膜尚光滑、完整,无核固缩表现,线粒体完整清晰。DCM组大鼠血清TNF-α、ROS显著高于NC组和Ket组(均P0.05)。DCM组大鼠心肌组织中肥大细胞分布密度显著高于NC组和Ket组。结论糖尿病大鼠心肌组织存在肥大细胞的表达增强,肥大细胞膜稳定剂酮替芬可减轻DCM。肥大细胞表达的增强可促进TNF-α和ROS的表达,部分参与DCM的发生发展。
[Abstract]:Objective to study the effect of ketotifen, a hypertrophic cell membrane stabilizer, on diabetic cardiomyopathy in rats and its possible mechanism. Methods 45 male Wistar rats were randomly divided into normal control group (NC group) and mast cell membrane stabilizer group (Ket group). DCM rat model was established by high fat and high glucose diet plus single intraperitoneal injection of streptozotocin (STZ). The Ket group was given 0.025% ketotifen solution 1 mg/kg intraperitoneal injection twice after the successful establishment of DCM rat model. At the same time, the NC group and the DCM group were given the same volume of normal saline blank control injection. After the experiment, all the rats were killed. The left ventricle was cut off, and the left ventricular weight was taken to calculate the whole heart index and the left ventricular mass index. The myocardial tissues of each group were observed and analyzed under electron microscope. The expression of tumor necrosis factor TNF- 伪 (Ros) in serum of rats in each group was detected by enzyme linked immunosorbent assay (Elisa). Results the total cardiac index in DCM group was significantly higher than that in NC group and Ket group (P 0.05). The left ventricular mass index in DCM group was significantly higher than that in NC group and Ket group (all P 0.05). The nucleus is clearly oval, The intercalated disc was continuous, the myofilament was dense, and a large number of mitochondria with normal morphology were found in DCM group. The myocardial nuclei of DCM group were constricted, mitochondria swelling, unclear, intercalated disc twisted and broken, and no obvious swelling of myocardial nucleus in Ket group. The nuclear film is still smooth, complete, nucleoless pyknosis, Compared with NC group and Ket group, the density of mast cells in myocardial tissue in DCM group was significantly higher than that in NC group and Ket group. Conclusion the expression of mast cells in myocardial tissue of diabetic rats is enhanced. Ketotifen, the stabilizer of mast cell membrane, can reduce the expression of DCM. The increased expression of mast cells can promote the expression of TNF- 伪 and ROS, and partly participate in the development of DCM.
