运动训练对高血压心脏功能的影响及其机制

【摘要】:目的:探讨运动训练对高血压大鼠的血压、心功能、心肌细胞收缩性、心肌细胞肌丝钙敏感性的影响。方法:大鼠随机分为四组,分别为高血压组,高血压运动组,假手术组,假手术运动组。利用血管紧张素II诱导高血压。多动物多通道尾袖套法无创血压系统,监测大鼠血压。心脏超声仪测左室射血分数。分离的心肌细胞,采用IonOptix单细胞收缩检测系统检测心肌细胞收缩性及心肌细胞肌丝钙敏感性。结果:(1)血压:运动2周,高血压组收缩压与高血压运动组收缩压有统计学意义(P0.05),高血压运动组的收缩压明显得到控制。假手术组收缩压与假手术运动组收缩压间无统计学意义(P0.05)。高血压组舒张压与高血压运动组舒张压间有统计学意义(P0.05),高血压运动组的舒张压明显得到控制。假手术组舒张压与假手术运动组舒张压间无统计学意义(P0.05);(2)左室射血分数(LVEF):运动训练后,高血压组LVEF与高血压运动组LVEF间有统计学意义(P0.05)。运动训练后大鼠LVEF有显著提高;(3)心肌细胞收缩性:运动训练后,高血压组心肌细胞收缩性与高血压运动组心肌细胞收缩性间有统计学意义(P0.05),运动训练后大鼠心肌收缩性明显恢复;(4)肌丝钙敏感性:运动训练使,高血压运动组肌节缩短长度与Ca2+浓度关系曲线向左移;运动训练后高血压组EC50与高血压运动组EC50间有统计学意义,(P<0.05),运动训练使EC50缩短。结论:1.运动训练能有效控制高血压大鼠的收缩压和舒张压;2.运动训练通过恢复心肌细胞肌丝Ca2+敏感性达到对心肌细胞收缩性及心功能的改善。
[Abstract]:Objective: to investigate the effects of exercise training on blood pressure, cardiac function, myocardial contractility and myofilm sensitivity in hypertensive rats. Methods: rats were randomly divided into four groups: hypertension group, hypertension exercise group and sham operation group. Sham operation exercise group. Hypertension was induced by angiotensin II. Non-invasive blood pressure system with multi-animal multi-channel tail sleeve method was used to monitor blood pressure in rats. Left ventricular ejection fraction (LVEF) was measured by cardiac ultrasound. Isolated cardiomyocytes were isolated. IonOptix single cell contraction detection system was used to detect the contractility of cardiomyocytes and the sensitivity of myofilm calcium. Results: blood pressure: exercise for 2 weeks. Systolic blood pressure in hypertension group was significantly higher than that in exercise group (P 0.05). Systolic blood pressure in exercise group was significantly controlled. There was no significant difference between systolic blood pressure in sham operation group and exercise group (P 0.05). There was significant difference between diastolic blood pressure (DBP) and diastolic blood pressure (DBP) in hypertensive exercise group (P 0.05). Diastolic blood pressure (DBP) in exercise group was significantly controlled. There was no significant difference between diastolic blood pressure (DBP) and diastolic blood pressure (DBP) in sham-operated exercise group (P < 0.05); left ventricular ejection fraction (LVEFV): after exercise training, There was significant difference between LVEF in hypertension group and LVEF in hypertensive exercise group (P 0.05). The contractility of cardiac myocytes in rats after exercise training was significantly increased after exercise training (P < 0.05). The contractility of cardiac myocytes in hypertension group was significantly different from that in exercise group (P 0.05). The contractility of myocardium recovered significantly after exercise training (P < 0.05). The relationship between the length of muscle ganglion shortening and the concentration of Ca2 in hypertensive exercise group was shifted to the left. After exercise training, there was significant difference between EC50 and EC50 in hypertension group (P < 0.05), and EC50 was shortened by exercise training. Conclusion 1. Exercise training can effectively control systolic and diastolic blood pressure in hypertensive rats. The sensitivity of myofilament Ca2 to cardiac contractility and cardiac function was improved.
