脂联素水平对心肌脂联素受体1表达的影响及其机制研究

【摘要】:目的:观察脂联素(adiponectin,APN)水平对心肌脂联素受体1(adiponectin receptors 1,AdipoR1)表达的影响;探讨胰岛素抵抗(insulin resistance,IR)是否是APN影响心肌AdipoR1表达的机制之一。方法:观察对象分为5组,其中野生型C57BL小鼠分为:对照组(CON组)和IR组,APN~(-/-)小鼠分为:APN~(-/-)组(KO组),APN~(-/-)+IR组(KIR组)和APN~(-/-)+IR+APN干预组(APN组)。IR组、KIR组、APN组小鼠给予高脂饲料诱导IR模型,其中APN组小鼠还给予重组APN腹腔注射。干预12周后取血,用血糖仪检测小鼠空腹血糖(fasting plasma glucose,FPG)、ELISA法检测血清空腹胰岛素(fasting insulin,FINS)及APN水平,并计算IR指数(HOMA-IR)。取小鼠心肌组织,采用免疫组织化学法、Western blot法、PCR法检测心肌AdipoR1的表达水平。结果:与CON组比较,KO组和IR组血清APN水平降低、心肌AdipoR1蛋白表达降低,IR组HOMA-IR水平升高。与KIR组比较,APN组血清APN水平升高、心肌AdipoR1蛋白升高、HOMA-IR水平降低。小鼠心肌AdipoR1与血清APN呈正相关,与FINS、HOMA-IR呈负相关。结论:APN可上调心肌AdipoR1的表达。IR可导致心肌AdipoR1表达下调。APN可通过改善IR起到上调心肌AdipoR1表达的作用。
[Abstract]:Objective: to observe the effect of adiponectin (APN) level on the expression of adiponectin receptors 1 (AdipoR1) in myocardium and whether insulin resistance is one of the mechanisms of APN affecting the expression of myocardial AdipoR1. Methods: the subjects were divided into 5 groups. The wild-type C57BL mice were divided into two groups: control group (Con group) and IR group (APN group) and IR group (n = 10). The mice in APN group were also given intraperitoneal injection of recombinant APN. After 12 weeks of intervention, blood samples were taken, fasting plasma glucosetins and APN levels were detected by Elisa, and the IR index was calculated. The expression of AdipoR1 in myocardium was detected by immunohistochemistry and Western blot. Results: compared with CON group, the serum APN level of KO group and IR group was lower than that of CON group. Compared with KIR group, compared with KIR group, serum APN level increased, myocardial AdipoR1 protein increased and HOMA-IR level decreased. There was a positive correlation between myocardial AdipoR1 and serum APN. Conclusion: AdipoR1 can up-regulate the expression of myocardial AdipoR1. Ir can induce the down-regulation of myocardial AdipoR1 expression. APN can up-regulate the expression of myocardial AdipoR1 by improving IR.
