【摘要】:基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)是基质金属蛋白酶大家族中的重要组成成员,属于明胶酶类,又称明胶酶B,具有降解与重塑细胞外基质(extracellular matrix,ECM)的作用。ECM又是心脏的重要组成部分,起支持和固定心脏的作用。正常情况下,ECM是相对稳定的。当心血管疾病发生时,MMP-9的表达量的变化使ECM的调节发生异常,使ECM的合成和降解失去平衡,引起ECM内成分的比例变化,影响ECM的正常的支持、信息传递等功能,使心脏表现出病理形态,失去心脏正常的生理功能,并产生多种并发症。在心脏中,ECM的降解对心脏重塑过程有重要影响,主要表现为细胞和心脏失去支持和固定,当心腔内压力升高时心肌细胞延伸,心腔扩大,心脏功能发生障碍,在心房中可以表现为心房壁变薄,心房扩大,正常的心房细胞成分减少,心房间质发生广泛纤维化,最终促进和维持心房重构的发生和发展。心房扩大和心房间质纤维化是心房结构重构的主要表现。由此可见MMP-9在心房重构中有重要作用。1 MMP-9不仅参与正常的生理过程如血管形成、创伤愈合、再生、胚胎发育等,而且在心血管疾病(如房颤、心力衰竭、二尖瓣疾病、心肌梗死等)发生、发展的病理过程中对心房结构重构也有重要影响。2通过抑制MMP-9的表达可以抑制心房重构,是治疗心血管疾病的一个重要方向。
[Abstract]:Matrix metalloproteinase-9 (MMP-9) is an important member of the matrix metalloproteinase-9 family. It belongs to gelatinases, also known as gelatinase B, and has the function of degradation and remodeling extracellular matrix metalloproteinase-9 (ECM). ECM is also an important part of the heart.To support and fix the heart.Under normal conditions, ECM is relatively stable.When cardiovascular disease occurs, the changes of MMP-9 expression make the regulation of ECM abnormal, make the synthesis and degradation of ECM out of balance, cause changes in the proportion of components in ECM, affect the normal support of ECM, information transmission and other functions.Make the heart show pathological form, lose the normal physiological function of the heart, and produce a variety of complications.The degradation of ECM in the heart plays an important role in the process of cardiac remodeling, which is characterized by the loss of support and fixation of cells and the heart.In the atrium, the atrial wall becomes thinner, the atrium expands, the normal atrial cell component decreases, the atrial interstitial fibrosis occurs, and the atrial remodeling is promoted and maintained.Atrial enlargement and atrial interstitial fibrosis are the main manifestations of atrial structural remodeling.It can be seen that MMP-9 plays an important role in atrial remodeling. 1 MMP-9 not only participates in normal physiological processes such as angiogenesis, wound healing, regeneration, embryonic development, but also in cardiovascular diseases (such as atrial fibrillation, heart failure, mitral valve disease, etc.)The occurrence and development of myocardial infarction have an important influence on atrial structural remodeling. 2 it can inhibit atrial remodeling by inhibiting the expression of MMP-9, which is an important direction in the treatment of cardiovascular diseases.
