非诺贝特对兔动脉粥样硬化斑块形成的影响及机制

【摘要】:目的探讨过氧化物酶体增殖物激活受体家族(PPARs)激动剂非诺贝特对动脉粥样硬化形成的影响及作用机制。方法将25只雄性新西兰白兔随机分为五组,每组5只,A组予标准饲料喂养,其余四组予高脂饲料喂养,均喂养8周。C、D、E组喂养4周结束时均行经股动脉球囊损伤术,并分别于手术当天给予生理盐水、非诺贝特、WY14643(非诺贝特酸的人工合成物)连续4周。喂养8周结束时,五组均进行股动脉内膜剥脱术,取股动脉,并留取血清。取各组股动脉标本,HE染色后显微镜下观察血管形态,计算血管狭窄率;采用免疫组化SP法及Western blotting法检测股动脉PPARα蛋白表达;采用RT-PCR法检测股动脉PPARαmRNA表达;采用ELISA法检测血清TNF-α、IL-10和P-选择素水平。结果 C、D、E组股动脉均有动脉硬化表现,D、E组表现均较C组轻,且E组较D组轻。与A组比较,其余四组管腔狭窄率均升高(P0.05或0.01);与C组比较,D、E组管腔狭窄率降低(P均0.05)。D、E组股动脉PPARα蛋白和mRNA表达均高于C组,血清IL-10水平高于C组,血清TNF-α和P-选择素水平均低于C组;组间比较P0.05或0.01。结论非诺贝特可减轻由高脂饲料和球囊损伤引起的兔动脉粥样硬化病变程度,增加IL-10表达、减少TNF-α和P-选择素表达,抑制炎症反应可能是其作用机制。
[Abstract]:Objective to investigate the effect and mechanism of fenofibrate, a peroxisome proliferator activated receptor family (PPARs) agonist, on atherosclerosis. Methods Twenty-five male New Zealand white rabbits were randomly divided into five groups. Five rabbits in each group were fed with standard diet and the other four groups were fed with high-fat diet. Normal saline and fenofibrate WY 14643 (artificial synthesis of fenofibrate) were given on the day of operation for 4 weeks. At the end of 8 weeks, the five groups were treated with femoral artery endarterectomy, femoral artery and serum. The vascular morphology was observed under microscope after HE staining, the expression of PPAR 伪 protein in femoral artery was detected by immunohistochemical SP method and Western blotting method, and the expression of PPAR 伪 mRNA in femoral artery was detected by RT-PCR. Serum levels of TNF- 伪 IL-10 and P- selectin were detected by Elisa. Results Arteriosclerosis was found in all femoral arteries in group C and group E was lighter than that in group D. Compared with group A, the rate of stenosis in the other four groups was significantly higher than that in group A (P0.05 or 0.01%), and the expression of PPAR 伪 protein and mRNA in femoral artery of group C was significantly higher than that in group C (P < 0.05), and the level of IL-10 in serum was higher than that in group C (P < 0.05), and the level of IL-10 in group E was higher than that in group C (P < 0.05). The serum levels of TNF- 伪 and P- selectin were lower than those of group C (P0.05 or 0.01). Conclusion fenofibrate can reduce the degree of atherosclerosis induced by high fat diet and balloon injury, increase the expression of IL-10, decrease the expression of TNF- 伪 and P- selectin, and inhibit the inflammatory reaction.
