AGT在再生障碍性贫血患者骨髓脂肪组织中的表达及其与血细胞的相关性分析

【摘要】:目的:本实验主要检测再生障碍性贫血(简称再障)患者骨髓脂肪组织中血管紧张素原(Angiotensinogen,AGT)的表达情况,并分析其对再障骨髓造血的影响。进一步探讨其在再障骨髓脂组织增多的病理发展进程中所起的作用,为研究再障脂肪组织增多的病理机制提供实验基础。方法:采用免疫组织化学技术(Immunohistochemistry,IHC)检测20例再障患者和20例骨髓象正常的白细胞减少症患者骨髓组织中AGT的表达,运用半定量分析软件检测AGT含量,并且分别对AGT与LYM、LYM%、NEU、HGB、PLT两变量进行相关性分析。结果:再障组血管紧张素原AGT表达水平(0.86±0.32)较对照组(0.23±0.16)明显升高,其中t=7.88,P0.001,差异有统计学意义。再障组AGT表达水平与患者LYM、LYM%呈现正相关关系(r分别为0.838、0.877,P均0.05),与患者NEU、HGB、PLT呈负相关关系(r分别为-0.667、-0.875、-0.860,P均0.05)。结论:AGT在再障患者骨髓脂肪组织中表达显著增加,且AGT与LYM、LYM%呈正相关关系,与NEU、HGB、PLT呈负相关关系。抑制再障患者脂肪组织AGT的表达可能为再障患者提供一个新的治疗方向。
[Abstract]:Objective: to investigate the expression of angiotensinogenin A GTT in bone marrow adipose tissue of patients with aplastic anemia (aplastic anemia), and to analyze its effect on bone marrow hematopoiesis of aplastic anemia. In order to provide experimental basis for studying the pathological mechanism of adipose tissue increase in aplastic anemia, this paper further discusses its role in the pathological development of adipose tissue in AA. Methods: immunohistochemical technique was used to detect the expression of AGT in 20 patients with aplastic anemia and 20 patients with leukopenia with normal bone marrow image. The content of AGT was detected by semi-quantitative analysis software. The correlation between AGT and LYMN HGBP PLT was analyzed. Results: the expression of angiotensinogen AGT in aplastic anemia group (0.86 卤0.32) was significantly higher than that in control group (0.23 卤0.16). There was a positive correlation between AGT expression level and LYMN LYM% in aplastic anemia group (P < 0. 05), and a negative correlation between AGT expression and HGBP (P = 0. 05) and 0. 05% -0. 667 (-0. 875) -0. 875- 0. 860 (P = 0. 05), respectively. Conclusion the expression of AGT in bone marrow adipose tissue of aplastic anemia patients was significantly increased, and there was a positive correlation between AGT and LYMU LYM%, and a negative correlation between the expression of AGT and neutrol HGBP PLT. Inhibition of AGT expression in adipose tissue may provide a new therapeutic direction for AA patients.
