Diplacone对同型半胱氨酸致血管内皮细胞损伤的保护作用

【摘要】:目的采用同型半胱氨酸(Hcy)孵育体外培养的人脐静脉内皮细胞(HUVEC),建立血管内皮细胞炎性损伤模型,后用Diplacone加以干预,观察Diplacone对HUVEC的影响,探讨Diplacone对血管内皮细胞可能的保护作用。方法先用不同浓度的Hcy预处理HUVEC,选出Hcy的最适损伤浓度(2 mmol/L)和时间(12 h),在HUVEC培养基中加入不同浓度Diplacone预处理2 h后加入2 mmol/L Hcy作用12 h,通过MTT法检测细胞活性变化,Hoechst33342核染色检测细胞核损伤程度,Western blot检测与细胞损伤有关的血管细胞黏附分子1(VCAM-1)和P-选择素的表达水平,以此衡量Diplacone对Hcy诱导的HUVEC损伤的保护作用。结果与对照组相比,2 mmol/L Hcy组细胞活性显著降低(P0.01),10μmol/L以内Diplacone具有明显的剂量依赖性抑制2 mmol/L Hcy诱导的HUVEC活性损伤并抑制Hcy所致的VCAM-1和P-选择素的表达升高(P0.01)。结论 Diplacone对Hcy所致的HUVEC内皮损伤具有保护作用。
[Abstract]:Objective to establish vascular endothelial cell inflammatory injury model by incubating human umbilical vein endothelial cells (HUVEC),) with homocysteine (Hcy) in vitro, and then to observe the effect of Diplacone on HUVEC by Diplacone intervention. To investigate the possible protective effect of Diplacone on vascular endothelial cells. Methods the optimal damage concentration (2 mmol/L) and time (12 h),) of Hcy were obtained by pretreatment of HUVEC, with different concentrations of Hcy for 2 h and 12 h after pretreatment with different concentrations of Diplacone in HUVEC medium. The changes of cell activity were detected by MTT and the expression of vascular cell adhesion molecule 1 (VCAM-1) and P- selectin were detected by Hoechst33342 nuclear staining, and the degree of nuclear damage was detected by, Western blot. The protective effect of Diplacone on HUVEC damage induced by Hcy was evaluated. Results compared with the control group, the cell activity of 2 mmol/L Hcy group was significantly decreased (P0.01). Within 10 渭 mol/L, Diplacone significantly inhibited HUVEC activity damage induced by 2 mmol/L Hcy and increased VCAM-1 and P- selectin expression induced by Hcy in a dose-dependent manner (P0.01). Conclusion Diplacone has protective effect on HUVEC endothelial injury induced by Hcy.
