注射用前列地尔及人参皂苷Rg3对大鼠心肌缺血/再灌注损伤的影响及机制研究

第一部分注射用前列地尔对大鼠心肌缺血/再灌注损伤的影响目的:观察注射用前列地尔对大鼠心肌缺血/再灌注损伤的影响,并初步探讨其作用机制。方法:(1)选用健康雄性SD大鼠,随机分为4组:假手术组(Sham组),心肌缺血/再灌注模型组(I/R组),前列地尔低剂量组(Alprostadil 4μg/kg),前列地尔高剂量组(Alprostadil 8μg/kg)。(2)结扎大鼠左冠状动脉前降支使其缺血30min,再灌注24h,制备心肌缺血/再灌注损伤模型。再灌注24h后检测血清肌钙蛋白T(Troponin T)、肌酸激酶同工酶(CK-MB)及乳酸脱氢酶(LDH)水平、NBT法检测心肌梗死面积、应用超声心动图检测大鼠心功能(再灌注72h)、在光镜及电镜下观察心肌组织形态学特点、TUNEL原位标记法和DAPI双重染色检测大鼠心肌细胞凋亡、检测心肌组织丙二醛(MDA)含量、超氧化物歧化酶(SOD)及过氧化氢酶(CAT)活性。结果:(1)与Sham组相比,模型组血清Troponin T、CK-MB及LDH均增高,差异有统计学意义(p<0.05)。与模型组相比,前列地尔4或8μg/kg组血清Tro...
