Cx43、E-cadherin、PI3K及CTGF蛋白在脉络膜黑色素瘤中表达的研究

【摘要】:目的探讨脉络膜黑色素瘤中侵袭与转移相关因子缝隙连接蛋白43(Cx43)、上皮型钙黏附分子(E-cadherin)、磷脂酰肌醇-3激酶(PI3K)和结缔组织生长因子(CTGF)的表达及意义。 方法实验研究。采用免疫组织化学法检测27例脉络膜黑色素瘤组织及15例色素痣组织中Cx43、E-cadherin、P13K和CTGF的表达,并与临床病理资料做对照分析,采用x2检验比较各因子表达阳性率的差异。 结果Cx43、E-cadherin、P13K及CTGF蛋白在脉络膜黑色素瘤中的表达率分别为74.07%(20/27),44.4%(12/27),74.07%(20/27)和66.67%(18/27),在色素痣中的表达率分别为33.33%(5/15),86.67%(13/15),33.33%(5/15)和20.00%(3/15),Cx43、P13K及CTGF蛋白在脉络膜黑色素瘤中的表达较色素痣增加,差异有统计学意义(x2=5.060,P=0.024;x2=5.060,P=0.024;x2=6.637,P=0.010)。E-cadherin蛋白在脉络膜黑色素瘤中的表达较色素痣降低,差异有统计学意义(x2=5.490,P=0.019)。Cx43蛋白在不同组织学类型的脉络膜黑色素瘤中阳性表达率不同:梭形细胞型为40%(4/10),混合型为88.89%(8/9),类上皮型为100%(8/8),差异有统计学意义(x2=9.874,P=0.007)。P13K蛋白在不同大小的脉络膜黑色素瘤中阳性表达率不同:小肿瘤为42.86%(3/7),中肿瘤为75%(9/12),大肿瘤为100%(8/8),差异有统计学意义(x2=6.357,P=0.042)。Cx43、E-cadherin、P13K及CTGF蛋白在未累及巩膜的脉络膜黑色素瘤中的表达率分别为50%(6/12),83.33%(10/12),50%(6/12)和41.66%(5/12),在累及巩膜的脉络膜黑色素瘤中的表达率分别为93.33%(14/15),40%(6/15),93.33%(14/15)和86.67%(13/15),Cx43、PI3K及CTGF蛋白在脉络膜黑色素瘤中的表达,累及巩膜组较未累及组增加,差异有统计学意义(x2=4.457,P=0.016;x2=4.457,P=0.016;x2=4.218,P=0.019)。E-cadherin在脉络膜黑色素瘤中的表达,累及巩膜组较未累及组降低,差异有统计学意义(x2=3.546,P=0.028)。 结论Cx43、P13K、CTGF蛋白的高表达及E-cadherin蛋白的低表达与脉络膜黑色素瘤的浸润和转移可能有密切的关系。
[Abstract]:Objective to investigate the expression and significance of invasion and metastasis related factor gap junction protein 43Cx43C, E-cadherinus, phosphatidylinositol 3 kinase (PI3K) and connective tissue growth factor (CTGF) in choroidal melanoma. Methods Experimental study was carried out. Immunohistochemical method was used to detect the expression of P13K and CTGF in 27 cases of choroidal melanoma and 15 cases of nevus pigmentosa. Results the expression rates of P13K and CTGF protein in choroidal melanoma were 74.07 and 74.44, respectively. The expression rates of P13K and CTGF protein in choroidal melanoma were 33. 33 / 515 / 86.67 and 33. 33% and 20 / 27, respectively. The expression rates of Cx43P13K and CTGF protein in choroidal melanoma were significantly higher than those in nevus of choroidal melanoma, and the expression of Cx433P13K and CTGF protein in choroidal melanoma was higher than that in nevus of choroidal melanoma, respectively. The expression rate of Cx43P13K and CTGF protein in choroidal melanoma was higher than that in nevus, and the expression rate of Cx43P13K and CTGF protein in choroidal melanoma was higher than that in nevus. The expression of E-cadherin protein in choroidal melanoma was significantly lower than that in nevus pigmentosa, and the expression of E-cadherin protein in choroidal melanoma was significantly lower than that in nevus pigmentosa. The difference was statistically significant in the positive rate of the protein in different histological types of choroidal melanoma: the spindle cell type was 40 / 10, the mixed type was 88.89 / 8 / 9, and the epithelioid type was 100 / 8 / 8 / 8. The difference was statistically significant (x29.874P0.007N. P13K protein) in different sizes of choroidal melanoma. The positive expression rates of Cx43E-cadherin P13K and CTGF protein in choroidal melanoma were 42.86%, 75 / 9 / 12 and 100 / 8 / 8, respectively. The difference was statistically significant. The expression rates of Cx43E-cadherin P13K and CTGF protein in non-scleral choroidal melanoma were 50 / 612 / 83.33 / 10 / 10 / 50 / 612) and 41.66% / 512P / r. The expression rates of Cx43 PI3K and CTGF protein in choroidal melanoma involving sclera were 93.3314 / 15, 93.3314 / 15, 93.3314 / 15 and 86.67%, respectively, and the expression of Cx43 PI3K and CTGF protein in choroidal melanoma. The expression of E-cadherin in choroidal melanoma was significantly higher in scleral group than that in non-involvement group, and the difference was statistically significant (P 0.016). The expression of E-cadherin in choroidal melanoma was significantly lower in scleral group than in non-involvement group, and the difference was statistically significant (P 0.02828) between the scleral group and the scleral group (P 0.016, P 0.016, P = 4.457, P = 0.021, P < 0.05), and the expression of E-cadherin in the scleral group was significantly lower than that in the sclera group, which was significantly lower than that in the sclera group. Conclusion the high expression of CTGF and the low expression of E-cadherin may be closely related to the invasion and metastasis of choroidal melanoma.
