谷氨酰胺诱导大鼠视网膜热休克蛋白70的表达

【摘要】:目的检测谷氨酰胺诱导HSP70在正常视网膜内的表达及随时间的变化。 方法健康清洁型Wistar大鼠90只随机分为三组,正常对照组10只、阳性对照组即硫酸锌腹腔注射组10只、实验组即谷氨酰胺腹腔注射组70只。实验组分别于谷氨酰胺注射(0.75 g/Kg)后1h,4h,6h,12h,24h,48h,72h各随机处死10只大鼠。阳性对照组于硫酸锌注射(24.6mg/Kg)后12h处死大鼠。正常对照组,阳性对照组及实验组各时间点10只老鼠中随机取5只用于免疫组织化学染色,剩余5只用于Western Blot分析。 结果Western Blot分析显示,正常对照组仅有HSP70的微量表达。谷氨酰胺注射1h后HSP70的表达稍有增强,4h后达到高峰并持续到12h,24h与48h组表达较前减少,但这两组之间无显著差异。72h组显著减少,但仍高于正常对照组。免疫组织化学染色显示诱导表达的HSP70在视网膜各层细胞均有明显表达,而正常对照组HSP70仅微量表达于视网膜内节层、外界膜及外核层。免疫组化染色阳性细胞光密度值测定结果与Western Blot结果一致。 结论1.正常情况下,视网膜微量表达HSP70 2.腹腔注射谷氨酰胺可以诱导大鼠视网膜HSP70的明显表达。
[Abstract]:Objective to detect the expression and changes of glutamine induced HSP70 in normal retina. Methods 90 healthy and clean Wistar rats were randomly divided into three groups: normal control group (n = 10), positive control group (n = 10) and glutamine intraperitoneal injection group (n = 70). In the experimental group, 10 rats were randomly killed at 1 h, 4 h, 6 h, 12 h, 24 h, 48 h and 72 h after injection of glutamine (0. 75 g/Kg). The rats in the positive control group were killed 12 hours after zinc sulfate injection (24.6mg/Kg). Five of the 10 mice in the control group, the positive control group and the experimental group were randomly selected for immunohistochemical staining, and the remaining 5 rats were used for Western Blot analysis. Results Western Blot analysis showed that there was only a trace expression of HSP70 in normal control group. The expression of HSP70 increased slightly after 1 h of glutamine injection, reached a peak at 4 h, and continued to decrease at 12 h and 48 h, but there was no significant difference between the two groups. The expression of HSP70 in 72 h group was significantly decreased, but still higher than that in normal control group. Immunohistochemical staining showed that the expression of HSP70 was significantly expressed in all layers of the retina, while in the normal control group, it was only expressed in the inner ganglion layer, outer membrane and outer nuclear layer of the retina. The results of optical density of positive cells by immunohistochemical staining were consistent with those of Western Blot. Conclusion 1. Under normal conditions, HSP70 2. 2 was expressed in retina. Intraperitoneal injection of glutamine could induce the expression of HSP70 in rat retina.
