正交试验优选虫草多糖脂质体的制备工艺

【摘要】:目的优选虫草多糖脂质体制备的最佳工艺。方法采用逆向蒸发法制备发酵虫草菌粉多糖脂质体,以包封率为指标,以药脂比、卵胆比与水油比为因素,采用L_9(3~4)正交试验设计对制备条件进行优选,鱼精蛋白测定发酵虫草菌粉多糖脂质体的包封率,透射电镜及马尔文激光粒度分析仪测定其形态和粒径。结果逆向蒸发法制备发酵虫草菌粉多糖脂质体的最佳工艺为大豆卵磷脂与药物的比例为20∶1,大豆卵磷脂与胆固醇的比例为2∶1,乙醚与磷酸盐缓冲液的比例为3∶1。结论采用最佳工艺制备的发酵虫草菌粉多糖脂质体包封率较高,形态和粒径较均匀,重现性好。
[Abstract]:Objective to select the best technology for preparation of polysaccharide liposomes from Cordyceps sinensis. Methods fermentation polysaccharide liposomes from Cordyceps sinensis were prepared by reverse evaporation. The encapsulation efficiency was taken as the index, the ratio of liposomes to liposomes, the ratio of egg to bile and the ratio of water to oil were used as factors. The preparation conditions were optimized by L _ (9) C _ (3) O _ (4) orthogonal design, and the entrapment efficiency of polysaccharide liposomes from fermented Cordyceps sinensis powder was determined by protamine. Results the optimum technology of preparing polysaccharide liposome from fermented Cordyceps sinensis powder by reverse evaporation was that the ratio of soybean lecithin to drug was 20: 1. The ratio of soybean lecithin to cholesterol was 2: 1, and the ratio of ether to phosphate buffer was 3: 1.Conclusion the entrapment efficiency of polysaccharide liposome from fermented Cordyceps sinensis powder prepared by the best technology is higher. The shape and particle size are uniform and the reproducibility is good.
