1株抗肿瘤银杏内生真菌的发酵条件及活性产物稳定性研究

【摘要】:对前期研究发现的1株分离自银杏树皮组织的具有良好的抗肿瘤活性内生真菌米曲霉菌(Aspergillus oryzae)YX-5菌株的发酵条件及活性产物稳定性进行研究。以发酵后的菌体干质量、粗提物质量和粗提物抗肿瘤活性为指标,对YX-5菌株的发酵温度、pH值、转速和发酵时间等条件进行优化。结果表明,最适宜的发酵条件为温度25℃、pH值7.0、摇床转速110 r/min、发酵时间7 d。以粗提物的抗肿瘤活性为指标,比较发酵液粗提物经热处理、酸碱处理、紫外照射和蛋白酶处理后的稳定性,发现YX-5菌株的活性产物在常温、中性和酸性环境中稳定性较好,对紫外线和蛋白酶有较强的抗性,但在超过100℃高温条件下或pH值超过9.0的碱性环境中稳定性较差。该菌株粗提物在自然条件下总体稳定性较高,不易发生变化,有利于活性成分分离工作的开展。
[Abstract]:The fermentation conditions and the stability of the active product of an endophytic fungus Aspergillus oryzae oryzae)YX-5, which was isolated from the bark of Ginkgo, were studied.
