醋酸曲普瑞林缓释微球体外加速释放及体内外相关性研究

【摘要】:目的建立体内外相关性良好的醋酸曲普瑞林缓释微球体外加速释放方法。方法残留法测定该药微球在大鼠体内的释放行为;分别考察表面活性剂、pH值、乙醇浓度、温度、转速和防腐剂浓度对体外加速释放的影响,调整释放条件并建立加速释放方法,进行体内外相关性研究。结果确定体外加速释放条件为:以15%乙醇溶液(含0.06%吐温80和0.1%苯扎氯铵)为释放介质,释放温度为55℃,振摇频率为200 r/min;在此条件下该药微球30 h释放达到87.35%,相当于微球在体内释放30 d,且体外加速释放结果与体内释放数据相关性良好(y=0.8845x+12.4510,R2=0.9938)。结论所建立的醋酸曲普瑞林微球体外加速释放方法与体内释放行为相关性良好,采用此法可快速有效评价该药微球释放行为。
[Abstract]:Objective to establish an accelerated release method of triprrelin acetate sustained release microspheres in vitro and in vivo. Methods the release behavior of the microspheres in rats was determined by residue method, and the effects of surfactants, pH value, ethanol concentration, temperature, rotating speed and preservative concentration on the accelerated release in vitro were investigated, the release conditions were adjusted and the accelerated release method was established to study the correlation in vitro and in vivo.
