药材甲热激蛋白基因SpHsp60的克隆及温度胁迫下的表达

【摘要】:本研究采用RT-PCR技术克隆了药材甲热激蛋白60(Heat shock protein 60,Hsp60)基因的cDNA全长序列,命名为SpHsp60(GenBank登录号:KX778623).氨基酸序列分析表明SpHsp60具有Hsp60基因家族高度保守的基序,实时定量PCR技术检测了不同温度胁迫后该基因的表达量,结果表明:-5℃和0℃低温处理2h,药材甲成虫体内的SpHsp60的表达量均显著高于对照组,且42℃高温处理2h后的表达量也显著高于对照组.说明SpHSP60与药材甲应对极端温度胁迫相关.
[Abstract]:In this study, the full-length cDNA sequence of heat shock protein 60 Hsp60 gene was cloned by RT-PCR and named SpHsp60 (GenBank accession number: KX778623). Amino acid sequence analysis showed that SpHsp60 had a highly conserved motif of Hsp60 gene family. The expression of SpHsp60 gene was detected by real-time quantitative PCR under different temperature stress. The results showed that the expression of SpHsp60 was significantly higher than that of the control group at the temperature of 5 鈩,
